| In this study,chicken breast cartilage was used as raw material to prepare chicken cartilage hydrolysate,collagen peptide and chondroitin sulfate,and its physicochemical properties were characterized.Based on the data,the effects of chicken breast cartilage hydrolysate,collagen peptide and chondroitin sulfate on osteoarthritis model rats were investigated by using sodium iodoacetate induced adult healthy male SD rats as osteoarthritis animal model.The main conclusions of this study are as follows:(1)The main components of dried cartilage were protein(71.74±0.36%)and carbohydrate(21.70±0.63%),followed by fat(0.24±0.03%)and ash(6.32±0.53%),indicating that chicken breast cartilage could be used to isolate collagen peptides and chondroitin sulfate.By calculation,the extraction rate of collagen peptide in chicken breast cartilage was 68.32±0.74%(measured by dry).the extraction rate of chondroitin sulfate was 18.82±0.09%(measured by dry).(2)The amino acid analysis showed that the content of glycine in collagen peptide was the highest,up to 14.05 mg/g,followed by glutamic acid(9.09 mg/g)and proline(7.44 mg/g)molecular weight of 92.38%collagen peptide was less than 1000 Da.Data analysis conducted by high performance liquid chromatography,fourier transform infrared spectroscopy,agarose gel electrophoresis and disaccharide composition analysis demonstrated that the extracted chondroitin sulfate was mainly composed of chondroitin sulfate A,with a relative purity of 91.87%.(3)In this study,sodium iodoacetate induced arthritis rats were used as a model.The results showed that the intervention of cartilage hydrolysate,collagen peptide and chondroitin sulfate could improve the symptoms of osteoarthritis induced by sodium iodoacetate and enhance exercise ability of arthritis rats.Through the detection of Tumor necrosis factor-α(TNF-α),interleukin-1 beta(IL-lβ),Prostaglandin E2(PGE-2),interleukin-6(IL-6)levels in rat serum,the results showed that gavage cartilage hydrolysate,collagen peptide and chondroitin sulfate could significantly reduce the TNF-α,IL-1β,PGE-2,IL-6 level in arthritis rats(p<0.05)and reduce the inflammatory response in arthritis rats compared with the model group.The results of histomorphology showed that the cartilage surface was intact after the intervention of cartilage hydrolysate,collagen peptide and chondroitin sulfate,the cell arrangement was regular,the number of cells increased obviously,and the gap between tissue and tidal line remained intact.The histopathological scores of cartilage hydrolysate group,collagen peptide group and chondroitin sulfate group were significantly lower than those of model group,indicating that the joint inflammation of rats was alleviated after intervention using cartilage hydrolysate,collagen peptide and chondroitin sulfate.The results of histomorphology suggested that the cartilage surface was intact after the intervention of cartilage hydrolysate,collagen peptide and chondroitin sulfate,the cell arrangement was regular,the number of cells increased obviously,and the gap between tissue and tidal line remained intact.The histopathological scores of cartilage hydrolysate group,collagen peptide group and chondroitin sulfate group were significantly lower than those of model group,which indicates that the joint inflammation of rats was alleviated after intervention using cartilage hydrolysate,collagen peptide and chondroitin sulfate. |
