Objective:By studying the effects of propofol pretreatment on Nrf-2,HO-1,Nox-1 protein expression,oxidative stress and inflammatory response in the ascending colon,to explore the effects of propofol pretreatment on the colonic ischemia-reperfusion injury in rats.Methods:Forty SD rats were randomly divided into sham operation group(Sham group,S group),ischemia reperfusion group(Ischemia reperfusion group,I/R group),and normal saline group(Normal saline group,NS Group),For three consecutive days before the operation,the rats in group P were given intraperitoneal injection of propofol 50mg/kg regularly,group NS was injected with the same dose of normal saline,group S and I/R were not treated,and the superior mesenteric artery was separated after opening the abdominal cavity in group S,but without Clamping,I/R group,NS group and P group was clamped the superior mesenteric artery for 1 hour and then perfused for 2 hours.After that,the ascending colon tissue was taken,and hematoxylin/eosin(H/E)staining was performed.The colon tissue was observed under light microscope for detecting Morphological changes and pathological damage scores,inflammatory factor content was measured:Interleukin-1,interleukin-6 and tumor necrosis factor-α contents in colon tissue were detected by enzyme-linked immunosorbent assay,detection of oxidative stress response:using xanthine oxidase method to measure superoxide dismutase activity and the thiobarbituric acid method was used to measure malondialdehyde content in colon tissue,detection of protein expression:Western blot was used to detect the expression of Nrf-2,HO-1 and Nox-1 proteins in colon tissue.Results:Compared with the S group,the pathological damage score of the colon tissue in the I/R group and the NS group were significantly increased(P<0.05),and the contents of IL-1,IL-6,TNF-α,and MDA were significantly increased,while the SOD activity was significantly reduced(P<0.05).Compared with the I/R group,the pathological damage score of the colon tissue in the P group was significantly reduced(P<0.05),The contents of IL-1,IL-6,TNF-α,and MDA were significantly reduced,while the activity of SOD in colon tissue were significantly increased(P<0.05).The expression of Nrf-2,HO-1 and Nox-1 protein in colon tissue of rats in group I/R and NS were significantly higher than those in group S(P<0.05);Compared with the I/R group and the NS group,the expression of Nrf-2 and HO-1 protein in the colon tissue of the P group increased,while the expression of Nox-1 protein decreased(P<0.05).Conclusion:Propofol pretreatment can increase the expression of Nrf-2 and HO-1 protein,while reduce the expression of Nox-1 protein and the content of tissue IL-1,IL-6,TNF-α and MDA at the same time,increase tissue SOD activity,reduce the degree of colonic pathological damage,It shows that propofol pretreatment has a protective effect on rats with colonic ischemia-reperfusion injury.
