Objective:To explore the effect of Sema4D expression on the immune function of CD8~+T cells in patients with chronic hepatitis B.Method:1.Collect 80 patients with chronic hepatitis B(CHB)as the test group and 30 healthy people as the control group.Collect the peripheral blood of the two groups of subjects and separate the serum.Ficoll density gradient centrifugation was used to separate peripheral blood mononuclear cells(PBMC),FCM was used to detect the expression of mSema4D on the surface of T cells in the two groups of PBMC,and the expression of s Sema4D in the serum of the two groups was detected by ELISA.The ICCS method combined with FCM was used to detect both the expression of IFN-γ,TNF-αand IL-2 in CD8~+T cells.2.The PBMCs of patients in the test group were divided into 3 groups.Group A was stimulated with HBV core antigen dominant epitope peptide(Core 18-27),and group B was stimulated with HBV surface antigen dominant epitope peptide(Env 183-191).In group C,complete medium was added.The expression of mSema4D on HBV specific CD8~+T cells and the expression of IFN-γ,TNF-αand IL-2 in CD8~+T cells were detected by ICCS combined with FCM.3.The level of ALT was detected by h7600 automatic biochemical analyzer,and HBV DNA was quantified by RT-PCR.The correlation of mSema4D expression with ALT level and viral load was analyzed.Results:1.Compared with the control group,the expression of mSema4D on peripheral blood T cells in the experimental group was increased(P<0.01),the expression of s Sema4D in plasma was decreased(P<0.05),and the expression of IFN-γ,TNF-αand IL-2 in CD8~+T cells was decreased(P<0.05).The difference between the above groups was statistically significant.2.In the experimental group,compared with group C,the expression of IFN-γ,TNF-αand IL-2 in CD8~+T cells of group A was significantly increased(P<0.05),while the expression of mSema4D on CD8~+T cells was significantly decreased(P<0.05),but there was no significant difference between group B and group C(P>0.05).3.The expression of mSema4D was positively correlated with ALT level and HBV DNA load(r_s=0.575,P<0.001;r_s=0.574,P<0.001).Conclusion:1.The high expression of mSema4D may be related to CD8~+0T cell immune dysfunction in patients with chronic hepatitis B.2.mSema4D can be used as one of the important indexes to monitor the immune function of chronic hepatitis B.3.Sema4D is expected to be the target of new antiviral drugs.
