Study On The Expression Of PFKFB3 In Osteosarcoma And Its Effect On The Proliferation And Metastasis Of Osteosarcoma Cells | | Posted on:2022-02-10 | Degree:Master | Type:Thesis | | Country:China | Candidate:L D Rao | Full Text:PDF | | GTID:2494306506979449 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Background and Objective:PFKFB3 is a key rate-limiting enzyme in the process of cellular glycolysis.It is widely involved in the molecular biological behavior of tumor cells and is closely related to the malignant growth and distant metastasis of tumors.This study aimed to analyze the expression level of PFKFB3 in osteosarcoma tissue and its effect on the proliferation and metastasis of osteosarcoma cells.Methods:1.Detect the expression of PFKFB3 in osteosarcoma tissue and its corresponding adjacent tissue samples by immunohistochemical staining,q RT-PCR and Western blot.2.Use sh RNA to silence the expression of PFKFB3 in osteosarcoma cells,Cell Counting Kit-8 and 5-ethynyl-29-deoxyuridine to detect the proliferation of osteosarcoma cells,and transwell and wound healing experiments to detect cells metastasis,flow cytometry to detect the proportion of apoptosis.3.In vivo experiments to detect the effect of PFKFB3 expression on the size of tumor-bearing mice and lung metastasis rate.Results:1.The results of immunohistochemical staining,q RT-PCR and Western blot confirmed that PFKFB3 m RNA and protein expression levels in osteosarcoma tissues were significantly increased(p<0.05).2.After down-regulating the expression of PFKFB3 in osteosarcoma cells,its proliferation ability was significantly inhibited(p<0.01),and the level of cell apoptosis increased(p<0.05).In the same way,the metastasis ability of osteosarcoma cells was significantly reduced(p<0.01)3.After down-regulating the expression of PFKFB3 in osteosarcoma cells,the volume and weight of tumor-bearing mice were significantly lower than those of the control group,and the incidence of lung metastasis was also lower than that of the normal control group.Conclusion:1.The expression of PFKFB3 in osteosarcoma tissues is significantly increased.2.Down-regulating the expression of PFKFB3 in osteosarcoma cells can significantly inhibit the proliferation and metastasis of osteosarcoma,and promote the occurrence of cell apoptosis.3.Animal experiments show that after inhibiting the expression of PFKFB3,its growth and lung metastasis are significantly inhibited. | | Keywords/Search Tags: | PFKFB3, Osteosarcoma, Proliferation, Migration, Invasion | PDF Full Text Request | Related items |
