The Effect Of MEBT/MEBO On STAT6 And SOCS1 Expression In Chronic Wound Tissue Repair

Objective: This study focused on the effect of MEBT / MEBO on SOCS1 and STAT6 in chronic wound healing.MEBT / MEBO here refers to skin regenerative medicine and therapy.It is hoped that the main research in this paper will be helpful for the promotion of MEBT /MEBO in the future,so that the relevant treatment methods can be carried forward.Methods:(1)150 Wistar rats were divided into model group,blank group,acute group,rb-b FGF group and MEBO group with 30 rats in each group.The wound healing of rats in each group was observed on the 3rd,7th and 14 th day after the experimental grouping,and the wound tissues were cut at these three time points for paraffin embedding,Masson staining and HE staining;(2)Western blot was used to detect and compare signal transducer and activator of transcription 6 and suppressor of cytokine signaling 1,and to analyze the protein expression of STAT6 and SOCS1 in wound tissue of rats in each group;(3)Real time PCR was used to detect the mRNA transcription of STAT6 and SOCS1 in granulation tissue of chronic refractory wound.Results:(1)By comparing the gross wound healing of each group and the results of Masson and HE staining of wound tissue,it was found that there was no significant difference between the MEBO group,the acute group and the rb-b FGF group,and the healing of these three groups was significantly better than that of the model group.(2)The expression levels of STAT6 and SOCS1 protein in wound tissue of each group were compared at each time point: the expression of STAT6 protein in acute group,MEBO group and rb-b FGF group decreased first and then increased(P < 0.05),while that in model group decreased continuously(P < 0.05).On the 3rd and 7th day after modeling,compared with the model group,the expression levels of STAT6 protein in the other four groups were lower.On the14 th day,there was no significant difference in the expression levels of STAT6 protein among the five groups(P > 0.05).The expression of SOCS1 protein in acute group,MEBO group and rb-b FGF group increased first and then decreased(P < 0.05),while that in model group increased continuously(P < 0.05);on the 3rd and 7th day after modeling,the expression of SOCS1 protein in acute group,MEBO group and rb-b FGF group was the highest,and that in model group was the lowest.On the 14 th day,there was no significant difference in the experimental results(P > 0.05).(3)Comparison of gene transcription levels of SOCS1 mRNA and STAT6 mRNA: after modeling,the transcription levels of STAT6 mRNA in acute group,MEBO group and rb-b FGF group showed a trend of first decrease and then increase(P <0.05),while that in model group showed a trend of continuous decrease(P < 0.05);on the 3rd and 7th day after modeling,the transcription levels of blank group,acute group,rb-b FGF group and MEBO group were significantly lower than those in model group(P < 0.05).There was no significant difference in mRNA transcription level among the five groups on the 14 th day(P > 0.05).After modeling,the transcription level of SOCS1 mRNA in acute group,MEBO group and rb-b FGF group increased first and then decreased(P < 0.05),and the model group increased continuously(P < 0.05);on the 3rd and 7th day after modeling,the transcription level of SOCS1 mRNA in acute group,rb-b FGF group and MEBO group was significantly higher than that in blank group and model group(P < 0.05),and the transcription level in blank group was higher than that in model group(P < 0.05).There was no significant difference in SOCS1 mRNA transcription between the five groups on the 14 th day.Conclusion:(1)The results of this study show that MEBT / MEBO is very worthy of promotion in the treatment of refractory wounds.It can not only reduce the infiltration of inflammatory cells,but also benefit the formation of cortical glands,hair follicles and capillaries.(2)The effect of this treatment is remarkable,which is mainly reflected in the regulation of STAT6 mRNA transcription level and STAT6 protein expression to achieve the inhibition of type 2 innate immunity,which is conducive to the realization of fibrosis and effective prevention of scar.(3)This treatment is beneficial to alleviate the inflammatory reaction and regenerate the wound tissue which is difficult to heal.
