Objective:To investigate the changes of neurotransmitters in cerebrospinal fluid caused by propofol,dexmedtomidine(Dexmedtomidine,Dex),ketamine(Ketamine,Ket)and cerebrospinal fluid during natural sleep,and identify different types of anesthetic sedatives Effects on neurotransmitters in cerebrospinal fluid.Methods: 32 healthy adult Sprague-Dawley(SD)male rats,weighing(300~350)g,were divided into 4 groups(n=8)using a random number table: control group: natural sleep group(Natural sleep,N Group);experimental group: propofol group,ketamine group,dexmedetomidine group.In the control group,rats were observed to sleep naturally during the day for 3 hours.Groups P,K,and D were intraperitoneally injected with propofol(100mg/Kg),ketamine(80mg/kg),and dexmedetomidine(100ug)./kg)until the rat falls asleep to a certain depth,and the rats are given intermittent administration to maintain the anesthesia state of the rat for 3h,and then the rats are anesthetized by intraperitoneal injection of 10% chloral hydrate(0.35ml/100g)in the four groups,and the rats are taken from the cerebellar cisterna Cerebrospinal fluid.Use enzyme-linked immunosorbent assay(ELISA)to detect norepinephrine(NE),dopamine(DA),serotonin(5-HT),acetylcholine(Ach),glutamate in cerebrospinal fluid The content of six neurotransmitters including glutamic acid(GLU)and gamma-aminobutyric acid(GABA).Using SPSS25.0 software,one-way analysis of variance was used to compare the obtained data between groups.Result:1.The content of GLU in the cerebrospinal fluid of group P,group K,and group D was significantly lower than that of group N(P<0.05),but there was no statistically significant difference in GLU content between group P,group K,and group D(P>0.05)).2.The content of GABA in the cerebrospinal fluid of group P,group K,and group D was significantly higher than that of group N(P<0.05),but the difference between group P,group K,and group D was not statistically significant(P>0.05)).3.The DA content in group P was significantly lower than that in group N(P<0.05),the content of DA in group K and D was significantly higher than that in group N(P<0.05),and the content of DA in group K was higher than that in group D(P<0.05)),The difference was statistically significant.4.The 5-HT concentration in group D was higher than that in group N(P<0.05),and the5-HT concentration in group P was lower than that in group N(P<0.05),and the differences were statistically significant.There was no significant difference in 5-HT content between group K and group N(P>0.05).5.The content of NE in group K was significantly higher than that of group N(P<0.05),the content of NE in group P and D was significantly lower than group N(P<0.05),and the content of NE in group D was lower than that of group P,the difference was statistically significant Academic significance(P<0.05).6.The Ach content of group D was higher than that of group N(P<0.05),and the Ach content of group P and K was lower than group N(P<0.05).The difference was statistically significant,but there was no statistically significant difference between group P and group K.Academic difference(P>0.05).Conclusion:1.Propofol,ketamine and dexmedetomidine may all increase the content of inhibitory neurotransmitter GABA and decrease the content of excitatory neurotransmitter GLU in cerebrospinal fluid,thereby exerting their anesthetic and sedative effects.2.Propofol can reduce the three monoamine neurotransmitters of NE,DA and 5-HT and Ach in the cerebrospinal fluid.Ketamine significantly increased NE and DA in cerebrospinal fluid,and decreased Ach,but had no effect on 5-HT.Dexmedetomidine increased 5-HT,DA,and Ach in cerebrospinal fluid,while NE significantly decreased.Compared with propofol and ketamine,the sedation induced by dexmedetomidine is easy to arouse and may bring about positive emotional reactions.
