Association Of TCF7L2 And SLC30A8 Genes Polymorphisms With Type 2 Diabetes Mellitus In Mongolian

Objective Genetic variation is a key factor in increasing the risk of diabetes.There are many relatively common type 2 diabetes mellitus single nucleotide polymorphism in the population,and each variation has a certain impact on the disease.In this study,we selected two candidate genes Transcription factor 7-like-2(TCF7L2)and Solute carrier family 30 member 8(SLC30A8)to evaluate their relationship with type 2 diabetes in Mongolian population.Methods In this study,patients who were treated in Inner Mongolia International Mongolia Hospital from March 2019 to June 2020 were entered into the case group and control group according to whether they had type 2 diabetes or not.The general data of sex and age of all subjects were obtained.Fasting blood glucose and blood lipids of five indexes were tested by automatic biochemical analyzer in the laboratory department of Inner Mongolia International Mongolian Hospital.The alleles and genotypes of TCF7L2(rs7903146)and SLC30A8(rs13266634)were encountered by a new generation of gene sequencing technique.Differences of indexes between the two groups were comparable by Student’s t-test.Allele frequencies of rs7903146 and rs13266634 were evaluated by chi-square goodness-of-fit test for Hardy-Weinberg.Finally,Logistic regression analysis was used to analyze the relationship between the variation of TCF7L2(rs7903146)and SLC30A8(rs 13266634)and type 2 diabetes with type 2 diabetes as dependent variables,alleles and genotypes as independent variables.Finally,the correlation between biochemical metabolic indexes and different genotypes of single nucleotide polymorphisms was analyzed by one-way analysis of variance.Results①A total of 246 Mongolian subjects were included in this study,including 148(95 males and 53 females)in the case group,with an average age of 60.87±10 years,and 98(42 males and 56 females)in the control group,with an average age of 45.24±15.19 years.Except fasting blood glucose,triglyceride and small and dense low density lipoprotein cholesterol(P<0.01),there were no significant differences in other biochemical indexes between the two groups(P>0.05).②The rs7903146 and rs13266634 in 246 subjects were in accordance with Hardy-Weinberg.③ T allele in the TCF7L2 gene is not linked with type 2 diabetes(OR=1.3455.95%CI=0.5917-3.059;p=0.4776).There was not any significant association between C allele in SLC30A8 gene and type 2 diabetes(OR=0.9015,95%Cl 0.6229-1.305;p=0.5823).④In five different genetic models,there was no significant association between rs7903146 and rsl3266634 and type 2 diabetes mellitus in Mongolian,excluding age and sex.⑤Mutations at the two sites were not associated with fasting blood glucose,triglycerides,and small and dense low-density lipoprotein cholesterol.Conclusion Polymorphisms of TCF7L2(rs7903146)and SLC30A8(rs13266634)genes may not be associated with type 2 diabetes in Mongolians,and research are needed to expand the sample size in the future.
