Objective To explore the relationship between single nucleotide polymorphisms of epidermal growth factor gene(EGF)sites rs4444903,rs11568835,rs2237051,rs2302135 and the risk of bladder cancer and kidney cancer in Chinese population as well as the interaction of environmental factors and polymorphisms at four sites on the risk of bladder cancer and kidney cancer.Methods The study used a 1:1 pair Case-control study method.We recruited 300 newly diagnosed bladder cancer patients and 152 renal cancer patients from the Department of Urology at the the Inner Mongolia and Inner Mongolia Medical College.The control groups come from healthy people undergoing physical examinations in the physical examination centers of these two hospitals in the sane team.The control groups and the case groups were matched 1:1 by the same sex and age (?) 5 years.A face-to-face questionnaire survey was conducted not only to collect their baseline information,including: general conditions,occupational exposure history,living and eating habits,but also collect blood samples,cancer and para-cancerous tissue samples from the subjects,and print their pathology reports.The multiple high-temperature ligase detection reaction technology(im LDR)was used to detect the genotypes of the four sites of EGF gene rs4444903,rs11568835,rs2237051,rs2302135.Enzyme-linked immunosorbent assay(ELISA)was used to determine the protein expression of EGF in serum.Real-time fluorescence quantitative PCR was used to determine the relative expression of m RNA in cancer tissues and adjacent tissues of EGF gene.All data are entered into Epidata3.1 and processed by IBM SPSS25.0 statistical software.If the measurement data obey the normal distribution,use "mean±standard deviation" to express,and use the "median(interquartile range)" for the non-normal distribution.For comparison between the two groups,if the variance is uniform,use the t test,and the variance Rank sum test is used for unevenness;enumeration data is expressed by rate or composition ratio,and comparison between two groups is used by X~2 test.The X~2 test was used to analyze whether the distribution of genotypes in the case group and the control group conformed to the Hardy-Weinberg equilibrium,and the genotype frequencies and alleles of EGF genes(rs4444903,rs11568835,rs2237051,and rs2302135)were analyzed the frequency distribution between the two groups.Binary logistic regression analysis was used to calculate the odds ratios(OR)of the co-dominant,dominant,and recessive genetic models;Multivariate conditional logistic regression was used to calculate the OR and the corresponding 95% confidence intervals(CI)of life and eating habits to the risk of bladder cancer and kidney cancer,and generalized multi-factor dimensionality reduction Method(GMDR)to analyze the interaction between genes and the environment.Results(1)Single factor analysis shows that compared with the control group,the bladder cancer case group has a history of education,pesticide exposure,factory work history,smoking,meat and vegetables,types of edible oil,fruit intake,drinking water type,There were statistically significant differences in hair dyeing,labor intensity,urine holding,heating and cooking(P<0.05);There were not statistically significant differences in BMI,drinking,dairy product intake,fried food intake,water intake,and air freshener use no statistical significance(P>0.05).Compared with the control group,the kidney cancer case group has statistical differences in education,pesticide exposure,dietary meat and vegetables,types of edible oil,fruit intake,dairy product intake,drinking water type,hair dyeing,labor intensity,heating and cooking Academic significance(P<0.05);There was no statistically significant difference in BMI,factory work history,smoking,alcohol consumption,fried food intake,water intake,urine retention,and air freshener use(P>0.05).(2)Multivariate logistic regression showed that exposure to pesticides,working in a factory,smoking,and heavy labor intensity were risk factors for bladder cancer(OR=3.732、2.239、2.498、2.103),a high education level is a protective factor for bladder cancer(OR=0.339).Exposure to pesticides,eating animal oil consumption and drinking deep well water is a risk factor for kidney cancer(OR=3.194、3.212、8.606),and high education and dairy product intake are protective factors for kidney cancer(OR =0.129、0.386).(3)The results of this study suggest that the three genetic models of codominance,dominant and recessive at EGF gene rs11568835,rs4444903,rs2237051,rs2302135 were not significantly associated with the incidence of bladder and kidney cancer(P>0.05).(4)The alleles and genotypes of the four loci of EGF gene were not significantly related to the pathological grade and clinical stage of bladder cancer(P>0.05).The genotype of rs2237051 locus was related to the pathological grade of renal cancer(P=0.029).(5)There were no statistically significant differences in the genotypes and allele frequencies of the four sites of EGF genes rs11568835,rs4444903,rs2237051,rs2302135 between the bladder cancer and kidney cancer case group and the control group(P>0.05).(6)GMDR analysis results showed that pesticide exposure history and factory work history are the dominant model for the interaction of bladder cancer(P=0.001);rs11568835,dietary meat and edible oil types are the dominant model for the interaction of kidney cancer(P=0.011).(7)The expression of EGF protein in serum of bladder cancer and kidney cancer was significantly higher than that in the control group(P<0.05).(8)The relative content of m RNA in the adjacent tissues of kidney cancer was significantly higher than that in kidney cancer tissues,and the difference was statistically significant(P<0.05).Conclusion(1)Exposure to pesticides,working in a factory,smoking,and heavy labor intensity were risk factors for bladder cancer,high education level is a protective factor for bladder cancer.Exposure to pesticides,eating animal oil consumption and drinking deep well water is a risk factor for kidney cancer;high education and dairy product intake are protective factors for kidney cancer.(2)The alleles and genotypes of the four sites of EGF gene have nothing to do with the pathological grade and clinical stage of bladder cancer;the genotype at the rs2237051 locus is related to the pathological grade of kidney cancer.(3)The results of this study suggest that the four sites of EGF gene rs11568835,rs4444903,rs2237051,rs2302135 are not related to the onset of bladder cancer and renal cancer under the three genetic models.(4)There is an interaction between the history of exposure to pesticides,and the history of factory work in the occurrence of bladder cancer;there is an interaction between rs2302135,edible oil type and fruit intake in the occurrence of renal cancer.(5)The expression of EGF protein in serum of bladder cancer and kidney cancer was significantly higher than that in the control group.(6)The relative content of m RNA in the adjacent tissues of kidney cancer was significantly higher than that in kidney cancer tissues.
