| Inflammatory bowel diseases,including Crohn’s disease and ulcerative colitis,are complex chronic recurrent diseases caused by multiple factors.Recently,the incidence rate has shown an upward trend,which greatly affects the quality of life of patients.So far,inflammatory bowel disease can not be cured yet.The clinical treatments of inflammatory bowel disease mainly focus on anti-inflammatory or immunosuppressive drugs.Although these therapies can temporarily relieve the symptoms,the recurrence of the disease cannot be prevented but with certain side effects.Therefore,new therapeutic strategies for inflammatory bowel disease still is imperative.Mesenchymal stem cells show potential for the treatment of inflammatory diseases through immune regulation and paracrine effect.The paracrine effect works by secreting soluble factors and releasing exosomes.After the exosomes reach the target cells and trigger signals through similar receptor-ligand interactions or transmit their contents(m RNA,mi RNA,enzymes,cytokines,chemokines and growth factors)through endocytosis.The therapeutic effect is similar to that of mesenchymal stem cells.Therefore,in addition to mesenchymal stem cells,exosomes provides a new strategy for the treatment of inflammatory bowel disease.The purpose of this study is to explore the function and internal mechanism of exosomes derived from mesenchymal stem cells of human perinatal tissue in the treatment of inflammatory bowel disease.In this study,we derived exosomes from human umbilical cord and placenta mesenchymal stem cell lines,generated dextran sodium sulfate induced inflammatory bowel diseases mouse model,and treated the mouse model by exosome injection through the tail vein.the histopathological HE staining was used to evaluated the therapeutic results.Further more,in vivo,immune factors,immune cells and instestinal microbiota before and after exosome treatment were examniated.Our results showed that 1.combined with colon length and histopathological HE staining,exosomes showed certain repair function on colon damage evaluated on the revovery of colon length and histopathology;2.exosomes promoted anti-inflammatory effect and regulate CD4~+T cells in vivo;3.exosomes can after exosome treatment,the intestinal flora of mouse models recovered to a level similar to that of control mice.The study will be benifical for the understanding the mechanism of mesenchymal stem cell and their exosomes on the treatment of inflammatory bowel disease and will play a role for the acceleration of clinical translation. |
