Objective: Rheumatoid arthritis(RA)is a systemic inflammatory disease,which can damage joints and synovium.The main pathological manifestations are chronic synovium inflammation,vascular inflammation and pannus formation.In the later stage of the disease,articular cartilage and bone will be destroyed,and eventually joints will become deformed and lose their normal function.This study comprehensively considered the effects of met + 2DG and MTX on ADMA level,aiming to find new inflammatory markers and new treatment methods for RA.Methods: 30 female Wistar rats were randomly divided into 6 groups: healthy control group,CIA group,CIA+MET+2DGH,CIA+MET+2DGM,CIA+MET+2DGL group and CIA+MTX group.The levels of ADMA protein in synovium and plasma of RA rats were measured by enzyme linked immunosorbent assay(ELISA).The expression levels of DDAH1 and DDAH2 in synovium and serum were measured by Western Blot method.The anti-inflammatory effect of each group of drugs on ankle joint of RA rats was evaluated by HE staining,and the relationship between the changes of ADMA,DDAH1 and DDAH2 levels and arthritis was analyzed.Results: 1.Inflammation: the average anti-inflammatory effect of CIA+MET+2DGH group was better than that of CIA+MTX group.The higher the dose of 2DG combined with MET,the better the anti-inflammatory effect.2.ADMA expression level: CIA+MET+2DGH group < CIA+MTX group <CIA+MET+2DGM group < CIA+MET+2DGL group.3.DDAH2 level: CIA+MET+2DGH group > CIA+MTX group > CIA+MET+2DGM group > CIA+MET+2DGL group.4.The level of DDAH1 has no obvious regularity.Conclusion: 1)the anti-inflammatory effect of MET+2DG combined with MTX is better than that of MTX,and the anti-inflammatory effect of MET+2DG is dose-dependent.2)MET+2DG and MTX promote the hydrolysis of ADMA by promoting the production of DDAH2.3)ADMA may be used as a new marker of synovitis in RA joint.
