| ObjectiveThe objective of this study was to detect the correlation between polycystic ovary syndrome(PCOS)and the single nucleotide gene polymorphism of Betatrophin and Apelin gene.Difference in the genotype of Betatrophin and Apelin between the PCOS patients and the control group was analyzed to investigate polymorphism of Betatrophin and Apelin polymorphism between the two groups.Based on previous studies,this study aimed at further exploring the etiology and pathogenesis of PCOS,identifying the risk factors for the occurrence and development of long-term complications in PCOS patients,thus to find gene sites for clinical diagnosis,treatment and prognosis of PCOS patients.Materials and Methods42 patients with PCOS who were treated in the Taishan Medicial University from January 2017 to October 2017 were selected as the subjects of the case group,and 44 infertility patients with problems of tubal factors,male factors or other factors were included in the control group during the same period,and the case-control study was carried out.The clinical basic indexes such as height,weight,body mass index,waist circumference,hip circumference,waist-hip ratio,fasting blood glucose,fasting insulin and serum lipid were measured,and the body mass index(BMI)was calculated.Then the patients were divided into different groups according to different markers,and the results were compared between the two groups and the subgroups.The genotype distribution of Betatrophin SNP(rs2278426)and Apelin SNPs(rs3115757,rs2235306 and rs3761581)were detected using Sanger method.The differences of genotype distribution were compared between the two groups to find the association of 4 SNP examined with the risk of PCOS.Results1.Among the 3 genotypes in Betatrophin gene locus rs2278426,there were 7 A/As,20 A/Gs and 15 G/Gs respectively in the PCOS group,and 4 A/As,13 A/G and 27 G/Gs respectively in the control group,and there were no statistical differences between the constituent ratios of the genotypes at this locus.But the frequency of A allele of rs2278426 was sigificantly higher in PCOS group than those in control group.2.Among the 3 genotypes in Apelin gene locus rs3115757,there were 5 G/Gs,16C/Gs and 21 C/Cs respectively in the PCOS group,and 11 G/Gs,20 C/Gs and 13 C/Cs respectively in the control group,and there were no statistical differences between the constituent ratios of the genotypes at this locus.And the frequency of C allele of rs3115757 were sigificantly higher in PCOS group than those in control group.3.Among the 3 genotypes in Betatrophin gene locus rs2235306,there are 12 T/Ts,16C/Ts and 14 C/Cs respectively in the PCOS group,and 19 T/Ts,18 C/Ts and 7 C/Cs respectively in the control group,and there were no statistical differences between the constituent ratios of the genotypes at this locus.However,the frequency of C allele of rs2235306 was sigificantly higher in PCOS group than those in control group.4.Among the 3 genotypes in Betatrophin gene locus rs3761581,there were 3 G/Gs,17 G/Ts and 22 T/Ts respectively in the PCOS group,and4 G/Gs,18 G/Ts and 22 T/Ts respectively in the control group,and there were no statistical differences between the constituent ratios and alleles of the genotypes at this locus.Conclusions1.The Betatrophin rs2278426 gene polymorphism maybe associated with PCOS of local women.The A allele may increase the risk of PCOS of women.2.The Apelin rs3115757,rs2235306 gene polymorphism maybe associated with PCOS of local women.The C allele may increase the risk of PCOS of women.3.The Apelin rs3761581 gene polymorphism may have no relation with PCOS of women. |
