Background: Osteoporosis(OP)is a common chronic bone disease.Menopausal women are more common.Due to the stealth and progressive nature of the disease,patients often pay attention to brittle fracture after serious complications,so early diagnosis,early treatment,early prevention of OP is particularly important.Exosomes have been shown to be involved in many aspects of bone metabolism and play an important role in the development of bone-related diseases.Mi RNAs,as important exocrine inclusions,play an important role in the early diagnosis,treatment and prognosis of osteoporosis.Based on RNA sequencing and biological information analysis,this study screened biomarkers for osteoporosis in postmenopausal women,and provided a new approach for early diagnosis and treatment of osteoporosis.Methods: From October 2019 to January 2020,12 osteoporosis patients(dual-energy X-ray: t =-2.5 or below)and 12 healthy controls were treated at the Anhui Medical University Lu’an Hospital.Serum samples were collected from osteoporosis patients and female healthy controls.RNA sequencing was used to analyze the amount of miRNA expression in plasma exocytosis and to identify the differentially expressed miRNA.Target Scan and miRanda database were used to screen and predict target genes.A Protein-protein interaction(PPI)network was constructed from a STRING database,and Cytoscape was used for data visualization and hub gene screening.Finally,Gene Ontology(GO)and KEGG(KEGG)were used to analyze the biological functions and signal pathways of target Genes.Results: A total of 1004 miRNAs were detected in 24 samples,and 11 significantly up-regulated miRNAs were found in postmenopausal osteoporosis women(p < 0.05),predicting 1968 differentially expressed target genes between different groups.According to the PPI network analysis,HDAC1,YWHAH,SOS1,YWHAZ,MED1,PIK3 CA,ITGB3,SMAD4,YWHAE and PDGFB were the 10 most significant genes in the PPI network.GO enrichment analysis showed that the target genes were mainly protein binding,metal ion binding,DNA binding,ATP binding and poly(a)RNA binding.PI3K-AKT signal pathway and mitogen-activated protein kinase(MAPK)signal pathway were the most enriched signal pathways.Conclusion: Serum exosome mirnas are associated with osteoporosis in postmenopausal women and may be a potential biomarker for predicting osteoporosis in postmenopausal women.
