Objective To investigate the role of Wnt/β-Catenin signaling pathway in the scar formation and the regulatory mechanism.Methods 1、The 21 patients(male 15 and female 6)underwent surgery in the Department of Orthopaedics,Affiliated Hospital of Qingdao University.Subcutaneous scar tissue and adjacent normal tissue are from operation incision of patients.2、Extraction of m RNA from tissue specimens.Real time Quantitative PCR technique was performed to detect the expression of Wnt/β-Catenin signaling pathway related genes,Axin-2、Dkk-1、c-Myc,in the fibroblasts,in order to understand and compare the activity of Wnt/β-Catenin signaling pathway in the scar tissue and adjacent normal tissue.3 、 Immunofluorescence technique was performed to detect the subcellular localization of β-Catenin.Results 1 、 The expression of Collagen-1 gene in fibroblasts of the scar tissue is obviously higher than that in normal tissue and the difference was significant(P < 0.005).Compared scar tissue and with normal tissue in the same individual,the expression of Axin-2、Dkk-1、c-Myc genes in fibroblasts of the scar tissue is higher than that in normal tissue,but their expression was not statistically difference(P>0.05).2、The distribution of β-Catenin in fibroblasts of the scar tissue is different from that in normal.β-Catenin is dispersed in the cytoplasm in fibroblasts of normal tissue,while in fibroblasts of the scar tissue,β-Catenin is accumulated adjacent the cell nucleus.Conclusions The subcellular localization of β-Catenin in fibroblasts of scar tissue is different from the fibroblasts of normal tissue:in the normal tissue,β-Catenin is dispersed in the cytoplasm,while in the scar tissue,β-Catenin gathered adjacent the cell nucleus.The activity of Wnt/β-Catenin in scar tissue and the surrounding normal tissue showed no significant difference.In the studies of model organisms,the activity of Wnt/β-Catenin signaling pathway was enhanced in the scar formation,but this phenomenon was not found on the study of the human scar tissue.It may have the following two explanations: First,in different model organisms,the activity of Wnt/β-Catenin signaling pathway did not change significantly in the process of the human scar formation;Secondly,because of the limitations of samples,we’ve got the scar tissue of mature scar tissue at 8 weeks after operation.In the process of scar formation,the adjustment process from activation to inhibit may be in Wnt/β-Catenin signaling pathway,the subcellular localization of β-Catenin may be involved in the regulation process. |
