Objective The research needs to determinine an ideal mouse model of immunological liver injury and apply the model to study on pharmacodynamics and mechanism of the water extract from Shan Li Huang Gen,which is a kind of She national medicine.Methods1.To distinguish the difference of immunological liver injury mouse model caused by ConA induced,BCG+LPS induced from observing serum liver enzymes ALT and drug AST,liver indices,etc from the researching,we want to select an appropriate animal modeling method for the following study2.Finally,we selected the BCG+LPS induced mouse model of immunological liver injury to study the affect liver function model and immune organs among the high,middle and low concentrations,which was extracted from Shan Li Huang Gen by water.Then,DDB was used as a positive control drug,by observing the spleen organ index,thymus index,serum ALT and AST liver drug enzyme liver pathology,to evaluate the drug efficacy3.We also selected the BCG+LPS induced mouse model of immunological liver injury to study the mice blood IFNlammatory mediators affected by giving the drug of Shan Li Huang Gen water extractor.DDB was used as a positive control drug while IFNlammatory mediators IL-2,IL-4,IL-10 and TNF-y were used to evaluate the drug efficacyResults1.Compared with the normal group,the hepatic metabolizing enzyme ALT,AST in the model of BCG+LPS mice were increased significantly,which the difference was statistically significant(p<0.05,n=10).Compared with normal group,the hepatic metabolizing enzyme ALT,AST in the model induced by Con A after the points of tail vein injection 8h,10h,12h,14,16h were all increased significantly,which the difference was statistically si gni fi cant(p<0.0 5,n=10)2.Shan Li Huang Gen high concentrations group effectively reduced the content of BCG+LPS-induced liver injury model in mice serum ALT and AST,compared with the model group,the difference was statistically significant(p<0.05,n=10),high concentration group also had the better efficiency than the DDS group at the down of liver serum ALT,the difference was statistically significant(p<0.05,n=10).Shan Li Huang Gen middle concentrations group reduced the AST level in blood only,compared with the model group,the difference was statistically significant(p<0.05,n=10),which was also better than the DDS group,the difference was statistically significant(p<0.05,n=10).Shan Li Huang Gen high concentrations group can effectively alleviate hepatomegaly caused by BCG+LPS-induced liver injury model.The difference of liver index compared with model group was statistically significant(p<0.05,n=10),while compared with the model group was not statistically significant(p>0.05,n=10).Liver tissue biopsy of Shan Li Huang Gen high concentrations group didn’t show any structural abnormalities while the middle group and the low group occasionally showed the spotty necrosis.Compared with the model group,the liver damage in these three treatment groups was significantly reduced3.Shan Li Huang Gen high and middle concentrations groups could effectively increase the level of mouse peripheral blood IL-2 and IL-10,compared with model group was statistically significant(p<0.05,n=10),while compared with the DDS group was not statistically significant(p>0.05,n=10).Shan Li Huang Gen high concentrations group could effectively decrease the level of mouse peripheral blood IL-4 and IFN-γ,compared with model group was statistically significant p<0.05,n=10),while compared with the DDS group was not statistically significant(p>0.05,n=10)Concludes1.All of the BCG+LPS-induced and ConA-induced immunological liver injury models can cause significantly liver damage,while the BCG+LPS induced mouse model has smaller group differences in ALT and AST than the ConA induced model2.Shan Li Huang Gen water extract can effectively reduce the BCG+LPS induced immunological liver injury in mice liver injury in lowering serum hepatic drug metabolizing enzyme ALT,AST,as well as reduce the liver index and improve on the liver injury pathological aspects,which the efficacy is the same as the drug of DDB3.By adjusting the release of the interleukins IL-2,IL-4,IL-10,IFN-γ,Shan Li Huang Gen water extract can regulate the Th1/Th2 balance to reduce the immunological liver injury in mice4.The water extract from Shan Li Huang Gen has a protective effect to immunological liver injury in mice.
