Objective:To observe the effect of Stigma maydis concentrated solution(SMC)on reducing blood glucose and blood lipids,and study the effect on blood sugar and fat metabolism of Stigma maydis polysaccharides(SMP)and Pu-erh tea polysaccharides.Methods:The extraction technology of Stigma maydis aqueous extract was optimized by the orthogonal test.The preparation of Stigma maydis polysaccharides(SMP)and Pu-erh tea polysaccharides were used ethanol subsiding method and freeze-drying technology.The content of polysaccharides in SMP and Pu-erh tea polysaccharides was measured by phenol-sulfuric acid method.The method for the assessment of assisting blood sugar reduction function of Technical Standards for Testing and Assessment of Health Food(2003)was used to observe the effect of SMC,SMP and Pu-erh tea polysaccharides on reducing blood glucose and blood lipids.Fasting blood glucose,total cholesterol,triglycetide,LDL-C and HDL-C were taken for test by the biochemical analyzer.The relative organ weight was measured by electronic analytical balance.To investigate the histopathological changes of the experimental rats by immunohistochemistry staining method.Results:The optimum extraction conditions for aqueous extract of Stigma maydis were as follows:solid-liquid ratio was 1/10,the extraction time was 4,the extraction time was 60 min;The polysaccharide content of SMP,ripened Pu-erh tea polysaccharides(RPP)and crude Pu-erh tea polysaccharides(CPP)were 27.58%,24.91%and 41.74%;In the study of SMC on blood sugar and fat metabolism,fasting glucose in middle dose SMC group,high dose SMC group and metformin group were lower than it in model group,with statistical significance(p<0.05).Total cholesterol in middle dose SMC group and high dose SMC group was declined significantly by comparison with model group(p<0.05),triglyceride in SMC group fell obviously with significant difference to model group(p<0.05),LDL-C in high dose SMC group was declined significantly by comparison with model group(p<0.05);In the comparative study of SMP RPP and CPP on blood sugar and fat metabolism,fasting glucose in small dose SMP group,middle dose SMP group,metformin group,high dose RPP group and CPP group were lower than it in model group,with statistical significance(p<0.05).Total cholesterol and triglycetide in SMP group and high dose RPP were declined significantly by comparison with model group(p<0.05),total cholesterol in small dose CPP group and middle dose CPP group was declined significantly by comparison with model group(p<0.05),LDL-C in high dose SMP group was declined significantly by comparison with model group(p<0.05),HDL-C in high dose SMP group and high dose CPP group rose obviously with significant difference to model group(p<0.05);The relative organ weight of liver in high dose SMC group was declined significantly by comparison with model group(p<0.05);The liver tissues of middle dose and high dose SMC group had less fat vacuoles than model group.Conclusions:SMC has effect of reducing blood glucose and blood lipids,SMC could prevent high fat diet on liver injury in rats.SMP,CPP and RPP are able to improve glucose and fat metabolism,CPP played the role of hypoglycemic effect more significantly and blood glucose decreased by approximately 40%of the model group.SMP played the role of hypolipidemic effect more significantly,total cholesterol,triglycetide and LDL-C decreased by approximately 25%,42%and 44%of the model group.
