The Study Of Continuous Waves EA Effect At Baihui(GV20),Shuigou(GV26)on Promoting The Permeation Of Rats’ Blood-Brain Barrier

Objective 1.To observe the effect of continuous waves EA at Baihui(GV20),Shuigou(GV26)on inducing EB to penetrate the rats’ Blood-Brain Barrier and to Select the better continuous wave frequency;2.To observe the effect of selected continuous waves EA at Baihui(GV20),Shuigou(GV26)on inducing NGF to penetrate the rats’ Blood-Brain Barrier.Methods 1.In the first part of experiment,the SD rat models of middle cerebral artery occlusion-reperfusion(MCAO/R)induced by Longa’s improved method were randomly divided into the model control group、2Hz group、15Hz group、30Hz group 50Hz group and 100Hz group after integrity of the blood-brain barrier in rats(21 days after operation),setting up another sham-operated group.The rats in each group were randomly divided into 2 subgroups to detect the brain water ratio and the penetration amounts of EB.The brain water ratio determination:Model control group and sham-operated group were fixed by cloth,without EA.EA groups were needled on Baihui(GV20),Shuigou(GV26)at corresponding frequency,2mA,for 8min.The rats were injected with anesthetics after intervention and perfused with saline through the left ventricle until the fluid flowing out of the right auricle turned colorless.Then,the right cerebrum was leaved without olfactory and cerebellum.The brain water ratio was examined by drying wet method.The penetration amounts of EB:after tail vein injection of 2%Evans blue(EB)saline solution(4mL/kg),model control group and sham-operated group were fixed by cloth,without EA.EA groups were needled on Baihui(GV20),Shuigou(GV26)at corresponding frequency,2mA,for 8min.The rats were injected with anesthetics after intervention.Perfusion saline through the left ventricle until the fluid flowing out of the right auricle turned colorless.Then,the right cerebrum was leaved without olfactory and cerebellum.The penetration of EB was examined by fluorescence method.2.In the second part of experiment,the SD rat models which were the same as the first experiment were randomly divided into model control group,2Hz group,100Hz group after integrity of the blood-brain barrier in rats(21 days after operation),setting up another normal control group.Three weeks later,after tail vein injection of NGF solution(NGF were diluted by PBS at 100ug/ml,injected at 10ug/kg),EA groups were needled on Baihui(GV20),Shuigou(GV26)at the corresponding frequency,2mA,for 8min.Normal control group and model control group were fixed by cloth,without EA.Then,brain tissues of all the groups were taken by perfusion.The NGF content was examined by ELISA.Results 1.The rat brain water ratio:there was no significant difference in all groups(P>0.05).2.The rat brain EB content:there was no significant difference between model control group and sham-operated group(P>0.05),while the rat brain EB content of 2Hz group and 100Hz group was significantly increased when compared with model control group,15Hz group,20Hz group and 50Hz group(P<0.05),there was no significant difference between 2Hz group and 100Hz group(P>0.05).3.The rat brain NGF content:there was no significant difference between normal control group and model control group(P>0.05).When compared with model control group,the rat brain NGF content of 2Hz group and 100Hz group was significantly increased(P<0.05),while the rat brain NGF content of 2Hz group was higher than that of 100Hz group(P<0.05).Conclusion 1.EB can be induced through the blood-brain barrier in rats and significantly increased by continuous wave EA at Baihui(GV20),Shuigou(GV26).Among that,the continuous waves with frequency 2Hz and 100Hz show the advantage,however they two have no difference from each other.In the meanwhile,different continuous waves EA on promoting the open of Blood-Brain Barrier won’t cause cerebral edema.2.NGF can be induced through the blood-brain barrier in rats by continuous wave 2Hz and 100Hz EA at Baihui(GV20),Shuigou(GV26).Among that,the continuous wave with frequency 2Hz shows the advantage.3.EA can induce NGF through the blood-brain barrier in rats,which provide an experimental foundation for the treatment of central nervous system diseases through the combination of acupuncture with medicine.
