Mechanism Of Jia-ji Electroacupuncture Combined With BWSTT On Axonal Regeneration In Spinal Cord Injury Rats Rho/ROCK Signaling Pathway

Objective To observe the expression of RhoA and neurite outgrowth inhibitor-A(Nogo-A),proteins in the pathway of Rho/ROCK,as well as the recovery of motive function in spinal cord injury rats under the treatment combined Jia-Ji electric-acupuncture(EA)with BWSTT.And to probe the mechanism of axonal regeneration in spinal cord injury rats under Jia-Ji EA combined with BWSTT on the basis of Rho/ROCK kinase signaling pathways.Methods 150 SD Male rats were randomly divided into sham group,model control group,exercise group,EA group,EA combined with exercise group,each group was randomly divided into three timing groups of 7th,14th and 28th day with 10 rats in each group.Acute moderate injury was caused by NYU instrument from the United States on the T9-T10 segment in modeling rats,while sham group only cut vertebral plate.After modeling,sham group and model control group were not treated,exercise group started body weight support treadmill training(BWSTT)at day 7 for 5 min and twice a day,EA group had Jia-ji acupuncture for treating around the damaged segment at the first day after modeling with 2/100Hz,1mA,daily,while EA combined with exercise group had a combination therapy of two intervention groups.Then had a BBB rating and killed for samples according to various time points.Immunohistochemistry and western blot were used to detect the expression of Nogo-A and RhoA.Results(1)the BBB rating,the first day after SCI,each modeling group had no significant difference,the scores at each group in the three time points increased to different extent,and the three intervention groups showed a trend of increaseing by degress that the difference is statistically significant(P<0.05).(2)Immunohistochemical,The results of NogoA showed that positive cells maintained in a low level in sham group,but increased to varying degrees in model groups.Three intervention groups compared with model group,the differences statistically significant(P<0.05),except the 7d in the exercise group.The two intervention groups compared with exercise group,the differences were statistically significant(P<0.05).Electric-acupuncture combined with exercise group compared with the pure electric-acupuncture group,the count of positive cells declined in certain degrees,the difference in 28 days group have statistical significance(P<0.05).As a result of RhoA,the count of sham group sustained in a stable low status at each time points.Compared with model group,the results of three intervention groups declined and the differences were statistically significant(P<0.05),except for the 7th day group in exercise group.The remaining two intervention groups compared with model group and exercise group,the positive cell count with a tendency of decline with statistically significant differences(P<0.05),28th day group excluded.Compared with the electric-acupuncture group,count of positive cells in electric-acupuncture combined with exercise group distinctly decreased,especially in the 14th,28th day groups,the differences statistically significant(P<0.05).(3)Western blot,It showed that the expression of NogoA in sham group keep in a low level,while higher in other groups.The results of each group were gradually decreased at three time points,compared with the model group,the intervention groups were significant differences except for the 7th day group in exercise group.Contrasted two intervention groups with exercise group,the outcome was significantly decreased(P<0.05).It declined both in EA combined exercise group and EA group but only in 28th day the contrast in two groups had a statistically significant difference(P<0.05).The expression of RhoA indicated that sham group remained at a low level status,high expression in SCI.The results in each group were decreased by degrees at three time points.Three intervention group compared with the model group,significant differences existed in each group excluding 7th day in exercise group.Compared with exercise group the other two intervention groups,the expression of interest protein in 28th day of EA group and all EA combined with BWSTT groups were significantly decreased(P<0.05).Contrasted EA combined exercise group with acupuncture group,the expression declined at the same time point,the differences of protein expression at the 28th day were statistically significant(P<0.05).Conclusions(1)Both Jia-ji EA and BWSTT(body weight support treadmill training)could contribute to the improvement of motor function in acute moderate spinal cord injury rats.(2)Jia-ji EA combined with BWSTT are superior to pure EA or BWSTT in the recovery of motor function in acute moderate SCI rats.(3)Jia-ji EA combined with BWSTT therapy may played a role in promoting axonal regeneration by Rho/ROCK signaling pathway.
