| Objective: To investgate the effect of TGP on proliferation and P38 MAPK signaling pathway in human rheumatoid arthritis fibroblast-like synoviocyte(HFLS-RA)induced by TNF-α.Methods : The HFLS-RA cell lines were recovered and subcultured to the 3-5generation.The HFLS-RA were stimulated with a final concentration of 10ng/ml TNF-α for 24 hours,the method of CCK8 was used to evalate the effect of TNF-α on proliferation in HFLS-RA.Then the HFLS-RA were stimulated with TNF-α after being intervened by different concentrations of TGP.And the effect of TGP on proliferation in TNF-α-induced HFLS-RA was examined by CCK8 assay.Western blot method was used to analysis the level of protein expression of the phosphorylated P38,MK2 and JNK in TNF-α-induced HFLS-RA intervened by different concentrations of TGP.Results: The CCK8 method showed that TNF-α significantly enhanced the proliferation of HLFS-RA,and the different concentrations of the TGP(2.5mg/ml,5.0mg/ml,7.5mg/ml,10mg/ml,12.5mg/ml)suppressed the proliferation of HLFS-RA induced by TNF-α in a dose-dependent manner.The Western blot method analysis showed that the protein expression of phosphorylated P38,MK2,JNK were significantly increased in TNF-α group.Howerever the different concentrations of the TGP reduced the protein expression of phosphorylated P38、MK2、JNK,and in a dose-depedent manner.Conclusions: TGP can inhibit the proliferation of HLFS-RA induced by TNF-α,and it can also reduce the protein expression of phosphorylated P38、MK2 and JNK.Hence,TGP exertes an anti-proliferative effect in FLS by inhibiting P38 MAPK kinase signaling pathway. |
