Discussion On The Mechanism Of "Protecting Bone" In Treating Rheumatoid Arthritis By Dkk/Wnt Signal Pathway

Objective:To observe the impact Qingre Huoxue Recipe on rats with rheumatoid arthritis in general morphology,pathological changes.Detecte the impact on related factors DKK1,β-catenin TCF,LRP5 of the Wnt signaling pathway.Investigate the effects of the bone protection mechanism on rats with rheumatoid arthritis cured with Qingre Huoxue Recipe.Methods:Use the CIA rat model,Divide the rat into normal group,Model control group,Qingre Huoxue Recipe treatment group and Etanercept treatment group.Investigate the effects on overall condition and joint swelling of the CIA rats.Observe the synovial and joint capsule pathology vanges of rats by synovial and joint capsule biopsy.Detecte the DKK-1,β-catenin mRNA expression level of knee joint capsule and synovium in every groups of rats by RT-PCR.Measure the serum levels of DKK-1,β-catenin,TCF,LRP5 protein levels by ELISA.Results:Qingre Huoxue Recipe relieve the degree of joint swelling and improve the overall situation of CIA rats.Synovial and joint capsule pathology biopsy display that inflammatory cells and synovial tissue of Qingre Huoxue Recipe treatment group rats less than the rats of model control group.Juxtaposed with the model group,DKK-1 gene expression level in joint capsule and synovium of Qingre Huoxue Recipe treatment group rats is lower than level in joint capsule and synovium of model control group rats(P<0.05).β-catenin gene expression was significantly up-regulated(P<0.05).Juxtaposed with the model group rats,DKK-1 content in the serum of Qingre Huoxue Recipe treatment group rats was significantly reduced(P<0.05).β-catenin,TCF,LRP5 content in the serum of Qingre Huoxue Recipe treatment group rats was significantly increased(P<0.05)Conclusion:Qingre Huoxue Recipe can improve the overall situation of CIA rats and relieve degree of joint swelling.Effective control the joint swelling inflammation symptom of most CIA rats.Reduce synovial proliferation and delay damaging effects of articular cartilage and bone erosion.By inhibiting the expression of Wnt signaling pathway DKK1 cytokines and promoting expression promoting ofβ-catenin cytokines,and then promote the formation of osteoblasts and play a role of bone protection.
