The Characteristics Of The Metabonomics And The Curative Effect Of Treatment By Chinese Herbs On The Basis Of The Rats’ Hyperuricemia

Objective:Build steady hyperuricemia model in rats by gavage with adenine to SD rat and hypodermic injection with Oteracil potassium,comparing Chinese and western medicine treatment of hyperuricemia tendency in the serum of the rats,and clear-cut Metabonomics characteristics of the hyperuricemia by treatment by chinese herbs.Methods:choose 100 healthy male SD rats,after adaptive feed 7 days,random divide into blank control group(A),Model group(B),Allopurinol+blank control group(C),Allopurinol+model group(D),High dose of traditional Chinese medicine+blank control group(E),High dose of traditional Chinese medicine+model group(F),medium dose of traditional Chinese medicine+blank control group(G),medium dose of traditional Chinese medicine+model group(H),low dose of traditional Chinese medicine+blank control group(I),low dose of traditional Chinese medicine+model group(J),10 rats in each group.all of blank control groups gavage with 25ml/kg distilled water In the morning,and inject with 4ml/kg sterile water in the abdominal skin twice(8 am,4 pm).all of model groups gavage with 100mg/kg adenine in the morning,and inject with 100mg/(kg·d)Oteracil potassium in the abdominal skin twice(8 am,4 pm).After the model building,A and B groups gavage with distilled water in the afternoon.C and D groups gavage with 10mg/kg Allopurinol in the afternoon.E and F groups gavage with 9.6g/kg.d traditional Chinese medicine named niaosuanlixian in the afternoon.G and H groups gavage with 4.8g/kg.d traditional Chinese medicine named niaosuanlixian in the afternoon.I and J groups gavage with 2.4g/kg.d traditional Chinese medicine named niaosuanlixian in the afternoon.liquid volume is 20ml/kg.After persist it for 14 days,narcotize rats with pentobarbital sodium.draw blood through Abdominal main vein for serologic tests and Metabonomics tests,and analyze it.pick liver and kidney,then observe its condition about the effect of Uric acid and drugs.Consequence:(1)condition of rats:Before the start of the experiment,all of the rats are active,hair luster,excrement and urine is normal.The weight of all rats have no difference(p>0.05).In the process of experiment,compared with blank control groups(A/C/E/G/I),rats of each model groups(B/D/F/H/J)are in bad mental condition,less activity,fur is unordered,diarrhoea.Fiinish the experiment,the weight of rats in each of blank control groups or in model groups have no difference(P>0.05).But compared with blank control groups,model groups is more light(P<0.05).After dissection,we find the kidney of rats of model groups.are damaged and the color turn gery.The liver of rats of each groups have no difference.(2)Uric acid:Compared with blank control group(A),the UA of model group(B)is increase(P<0.05).Compared with model group(B),Allopurinol+model group(D)is less about UA.Traditional Chinese medicine+model group(F/H/J)are also less about UA,and high dose of traditional Chinese medicine+model group(F)is least(P<0.05).Compared with Allopurinol+model group(D),high dose of traditional Chinese medicine+model group(F)has no difference.Creatinine and urea:Compared with blank control group(A),model group is higher about Cr and Urea(P<0.05).Compared with model group,allopurinol+model group(D)has no difference about Urea,but is less about Cr(P<0.05).High and medium dose of traditional Chinese medicine+model groups(F/H)are less about Cr and Urea.And low dose of traditional Chinese medicine+model group(J)has no difference(p>0.05).Alkaline Phosphatase:Compared with model group(B),traditional Chinese medicine+model groups(F/H/J)are less about alkaline phosphatase,but have no significance statistics(p>0.05).Allopurinol+model group(D)is Increase in trend(p>0.05).Compared with allopurinol+model group(D),traditional Chinese medicine+model groups(F/H/J)are less about alkaline phosphatase(P<0.05).Low density lipoprotein:compared with model group(B),traditional Chinese medicine+model groups(F/H/J)are less about LDL(P<0.05).(3)Through the analysis of metabonomics about allopurinol+model group(D)and high dose of traditional Chinese medicine+model group(F),wo fing that,retention time 5-11 min,the major compound is Urea.Retention time 11-16min,the major compound are Aspartic acid,Alpha-Ketoglutaric acid,Proline,Glycerol-3-phospate,Glutamine.Retention time 17-30min,the major compound are 9-(Z,Z)-octadecadienoic acid,9-(Z)-octadecadienoic acid,Cholesterol.Conclusion:(1)Through gavage with adenine and injection with Oteracil potassium can Build steady hyperuricemia model similar with human body.(2)Different dose of traditional Chinese medicine have difference not only in decreasing serum uric acid,but also in protecting kidney and decreasing LDL.High dose of traditional Chinese medicine is best.And it maybe protect liver.(3)Through metabonomics,we find traditional Chinese medicine can promote excretion about Urea,amino acid and fatty acid when treat hyperuricemia.
