Study On The Compounds Of Cordyceps Cicadae And Coydalis Decumbens(Thunb.)Pers.

Objectives:Cordyceps cicadae is the dried complex bodies of cicada which is parasited by Paecilomyces cicadae,Coydalis decumbens(Thunb.)Pers.is the dried tuber of Corydalis decumbens.Systematical investigations on the chemical constitutes of the TCMs,Cordyceps cicadae and Coydalis decumbens(Thunb.)Pers.were carried out.We expected to obtain new compounds and provided certain experimental basis for further exploitation of the TCMs.Methods:①The pulverized Cordyceps cicadae were extracted by 95%ethanol to make crude extract,then the crude product was extracted by petroleum ether,ethyl acetate and n-BuOH successively to get each position.Next the each position was purified by silica gel,reverse phase silica gel and LH-20 gel to get individual compound.② The pulverized Coydalis decumbens(Thunb.)Pers.were extracted by 95%ethanol to make crude extract.Total alkaloids were separated from the crude extract according to the acid soluble alkali sink theory.Next the total alkaloids were purified by silica gel,reverse phase silica gel and LH-20 gel to get individual compound.③Each compound was identified by simple contrast,HPLC and spectroscopic methods.The structures of new compounds were determined mainly by MS,high resolution MS,1D spectrogram(1H-NMR/13C-NMR/DEPT)and 2D spectrogram(HSQC/HMC/COSY/NOESY).Results:①20 compounds were isolated when we studied on the chemical components of Cordyceps cicadae.Among those compounds,one compound,identified as 5,5’-bis((2-ethylh-exyl)oxy)-2,2’-bifuran,was totally a new finding,and 2 compounds,identified as(2R,3S)-2-(3’,4’-dihydroxy-phenyl)-3-acetylamino-7-(N-acetyl-2"-amino-ethylene)-1,4-benzodixane,((2,3,4-trihydroxy-5-(hydroxymethyl)tetrahydrofuran-2-yl)-methyl)tryptophan,were isolated from this TCM for the first time.The rest were known compounds,They were respectively N-Acetylnoradrenoline,N-(2-hydroxyethyl)-adenosine,Ergosterol,2-Elaidoylglycerol,Elaidic acid,Cytosine,Uridine,2’-Deoxy-uridine,Thymidine,Adenosine,2’-Deoxy-adenosine,Isoguanosine,2’-Deoxy-isoguanosine,Cordycepin,Tyrosine,PHE,Tryptophan.②In the study of the components of Coydalis decumbens(Thunb.)Pers.totally 5 compounds were determined,including 2 new compounds,identified as Decumbensine and Decumbendine,and 3 known compounds,identified as Tetrahydropalmatine,5,6-dihydro-2,3,9,10-di(methylenedioxy)-8H-dibenzo[a,g]quinolizin-8one and Protopine.Conclutions:We made new findings through various identification methods when studying the compounds of two TCMs,Cordyceps cicadae and Coydalis decumbens(Thunb.)Pers.and enlarged the compounds library of two TCMs.
