Purpose:Observed pirfenidone sustained-release film delivery system to inhibit scarring of filtering channel after glaucoma filtration surgery in the rabbit eye.Methods:Trabeculectomy were performed on 48 eyes of 48 rabbits which were divided into 4 groups,group A(12 rabbits)was treated with the pirfenidone film ofatrigel under sclerotic flap,group B(12 rabbits)was treated with trabecu lectomy plus implantion of chitosan film under sclerotic flap,group C(12 rabbits)was performed trabeculectomy using MMC(0.25mg/ml)under sclerotic flap,group D(12 rabbits)played the role of control which had surgery only.For the right eye surgery.Postoperative intraocular pressure(IOP),inflammation,conjunctival bleb,and random selection of experimental animals,respectively,after 7,days 14,days 21,days 28 days to take the aqueous humor line ELISA test TGF-β2 factor in expression,respectively after 7 days,14 days,28 days randomly selected experimental animals were sacrificed one,removal of eye surgery eye histological examination.Results:Animal experiments showed that the longest of the postoperative group A,C group buck,followed by the B group,group D to maintain buck the shortest time;A set of functional blebs relatively prolonged survival time compared with the other three groups of functional blebs,moreover,conjunctival filtration taking a dip in blood vessels,pale;A group of postoperative complications compared with the other three groups,no increase;Pathology revealed A group 4 weeks after filtration canal,the degree of scarring is minor compared with the other groups;ELISA for detection of group A after 1 week in aqueous humor TGF-β2 content in significantly reduced,the other three groups in the aqueous humor TGF-β2 content increased.Conclusion:The pirfenidone sustained-release film delivery system used in glaucoma filtration surgery histocompatibility,inhibit scarring effect in the short term,be able to effectively maintain the filtration,the IOP is satisfied,no serious complications occurred,safe and reliable.Need to extend the stability of the release time of the sustained-release film delivery system,maintain an effective release of the concentration,shorten the degradation time,wound repair cycle synchronization,better inhibition of scarring.
