Objective:To evaluate the in vitro and in vivo effects of Photolon mediated photodynamic therapy(PDT)on Hela human cervix cancer cells.Methods:The cells were co-incubated with a series concentration(0.125-2.0ug/ml)of Photolon for 12 hours and irradiated with a light-fluence of 2.5 J/cm2 for 1,2,3,and 4 days,respectively.Cell viability was evaluated by MTT assay.The cells were treated with photolon at different doses(0.25ug/ml,0.5ug/ml)for 3 hours and then radiated by laser with energy scopes.The morphological characteristics were observed by light microscopy.The in vivo efficacy of Photolon mediated PDT(photolon-PDT)was assessed using a xenograft tumor model subcutaneously established in BALB/c nude mice.For a histologic analysis,hematoxylin and eosin staining was performed.Results:1.The photolon(0.25ug/ml)-PDT significantly reduced cell viability of Hela cells in a time-dependent manner.Compared with the control group,the cell proliferation inhibition rate were 26.87%,15.08%,11.99%,and 14.81%,respectively(P<0.05).2.Photolon-PDT significantly inhibited the growth of Hela cells in xenograft models.After 5,7,9,11,14,16,18,21,23,25,28 days treatment.The tumor inhibited rate was 43.73%,41.82%,49.43%,45.90%,47.97%,54.23%,54.25%,53.35%,58.49%,64.76%;and 68.23%,respectively(P<0.05).3.The Photolon was accumulated in the cytoplasm of the Hela cells,but not in the nucleus.4.Typical apoptosis morphological features cell shrinkage and condensation and margination of nuclear chromatin were observed after Photolon-PDT.Conclusion:Photolon-PDT inhibits the growth of Hela cells dependent on the Photolon concentration and laser exposure time in vitro and in vivo,indicating that Photolon should be considered a potent photosensitizer for cervical carcinoma treatment.
