ObJective:The flower of Edgeworthia gardneri Wall.Meisn.,named "Lv-Luo-Hua" in Chinese which belongs to genus Edgeworthia in Thymelaeaceae family is peculiar in Tibet.Through the in vitro phar macodynamics experiment to determine the effective part of the flower hypoglycemic effectIsolate th e identifed compounds from the active components.Evaluate the hypoglycemic activity of the Edgew orthia gardneri Wall.Meisn.and quality controlof the flower herbs.Methods:In this experiment,the dried power of the flower of Edgeworthia gardneri was extracted t hree times with 70%ethanol for 3 times.The combined extracts were concentrated under reduced pr essure,and the residue was partitioned into water and extracted with petroleum ether,EtOAc,and n-BuOH,successively.α-Glucosidase enzyme inhibition in vitro experiment is adopted to define the act ive site of the flower.Open positive phase silica gel column chromatography,polydextran gel,preparat ion high performance liquid chromatography and recrystallization methods was used to isolated comp ounds from Water insoluble part and Ethyl acetate extraction.The constructure of the compounds wa s determined by physical,chemical and the modern spectrum technology(1H-NMR,3C-NMR,2D-N MR,ESI-MS).Thecompounds were evaluated in vitro of hypoglycemic activity.The content determina tion method of kaempferol-3-O-β-D-(6’-O-trans-coumaroyl)-glycopyranoside were established by HPLC.CONCLUSION:The activity in vitro studies have shown that the flower for α-glucosidase enzymes has a certain inhibitory effect,and the ethyl acetate extract had the strongest activity.The water-inso luble fraction of the extract and the ethyl acetate extract fraction were systematically separated,seven compounds were isolated and identified Thier are kaempferol-3-O-β-D-(6’-O-trams-coumaroyl)-glycopy ranoside(1),8-(3-(2,4-benzenediol)-propionic acid methyl ester)-coumarin-7-β-D-glucoside(3),4-(4-hydr oxy-3,5-dimethoxyphenyl)-3-(hydroxymethyl)butane-1,2,4-triol(5),4-(4-hydroxy-3-methoxyphenyl)-3-(hydr oxymethyl)butane-1,2,4-t riol(6),p-carboxyphenyl β-D-glycopyranoside(7),edgeworoside C(8).The com pound(2)and the compound(4)were insolubled in any reagent and the structure needs to be furt her identified The α-Glucosidase activity assary showed thatethyl acetate extract in concentration in t he 5 μg/ml~160 μg/ml,the inhibition rate was 5.96%~69.6%.Compond(1)in concentration in the 5μg/ml~160 μg/ml,the inhibition rate was 5.03%~22.59%.The content determination methods of kae mpferol-3-O-β-D-(6’-O-trans-coumaroyl)-glycopyranoside(1)was determined.The results show that tilir oside in 0.105~1.47 mg had a good linear relationship with absorbance in the range.And the conte nt of kaempferol-3-O-β-D-(6’-O-trans-coumaroyl)-glycopyranoside(1)in the flower of Edgeworthia gar dneri was 1.2%.
