Objective1.To fabricate an artificial nerve graft by using collagen as bioactive component material to simulate the bionic structure of nerve.2.To verify the formation of combinatorial different forms of collagen nerve conduit can improve the repair of nerve injury.Methods1.Use acetic acid to isollated collagen from rat tails,and obtain different collagen hydrogel solution with concentration of 90 mg/ml,20 mg/ml,1 mg/ml.Use the freeze drying method to process collagen.Obtain different forms of collagen.The collagen films was used to simulate the epineurium,the multi tunnel collagen scaffolds was used to simulate the perineurium and the collagen hydrogel was used to simulate the extracellular matrix(ECM)and the endoneurium.The morphological and mechanical properties of the scaffolds were observed by Scanning Electron Microscopy.2.Assemble these collagen graft components together as an biomimetic artificial nerve graft to bridge the 10 mm long sciatic nerve defect model in rats.With the allograft nerve(PN)as control,using behavioral tests,electrophysiological and histomorphological analysis to detection the effect of this artificial nerve graft in the repair of peripheral nerve injury.Results1.The multi tunnel scaffold revealed that the aligned scaffolds had a highly porous interior.These porous were approximately 15-55 μM in diameter as aligned topographical features on the scaffold,which would guide cell behavior.And the scaffold has a degree of porosity,swelling rate and mechanical,which is good for the cell growth in the early stage after transplantation.2.The biomimetic artificial collagen nerve conduit obtained by compositing the outer tubular collagen conduit,the multi tunnel collagen scaffolds and collagen hydrogel solution,can effectively promote the regeneration of sciatic nerve defects in rats.It can promote axonal regeneration,myelination,distal target organ,nerve conduction function and motor function recovery after nerve injury.Conclusions1.Using different concentrations of collagen hydrogel to make the scaffold to simulate the epineurium and perin eurium and endoneurium,can provide a good raw material for bionic nerve graft and a new production process strategy for tissue engineering.2.The biomimetic artificial collagen nerve conduit obtained by compositing the outer tubular collagen conduit,the multi tunnel collagen scaffolds and collagen hydrogel solution,can effectively promote the regeneration of sciatic nerve defects in rats.
