ObjectiveThrough the observation of dioscore porridge before and after the intervention of type 2 diabetes mellitus model(T2DM)SCAFs in intestinal tract of rats(acetic acid,propionic acid and butyric acid)and lipopolysaccharide(LPS),the changes of indexes of Bifidobacterium total bacteria count and blood sugar in the study of dioscorea porridge on T2DM rat intestinal flora metabolism influence for dioscore porridge continue research and application to provide a scientific basis for.Methods40 male Wistar rats were selected and 8 were randomly selected as the blank group and the modeling group.They were fed with common feed and high sugar and high fat diet,respectively.6 weeks later,the fasting could not help the water 12h overnight.The model group was injected with 1%streptozotocin(STZ)citric acid buffer at a dose of 25mg/kg,and the same dose was repeated once after 72 hours.After the two injection of 72h,FBG was measured by the caudal vein,with the symptoms of two times FBG more than 11.lmmol/L in the same day,with multiple drinks,polypodities,polyuria,weight loss and so on.It was regarded as the success index of the model of T2D M rats.In this experiment,32 rats were made and the models were successfully made.After the model was successful,each group was randomly divided into model group and Dioscorea congee group(Dioscorea group and Dioscorea congee group).Group 8 rats.The blank group and the model group were intragastric 15ml/kg 1 times a day at 9:00,and the metformin group was gavage 1 times a day at 9:00 a.m.dimethyl metformin aqueous solution 15ml/kg,and the Dioscorea congee 15m1/kg was gavage 1 times a day in the Dioscorea congee group,and the intervention was 6 weeks for a long time.During the intervention period,the body weight and FBG were measured weekly.After 6 weeks of intervention,the concentration of SCAFs(acetic acid,propionic acid and butyric acid)in the colon content of rats was detected by gas chromatography mass spectrometry(GC-MS),and the level of LPS in serum was detected by ELISA,and the content of Bifidobacterium in the colon content of rats was detected by RT-PCR.Results1.Comparative analysis of FBG in rats of each group after interventionThe FBG of T2DM model rats fed with high glucose and high fat diet for 6 weeks combined with small dose of STZ induced model rats,the FBG of each model rat was greater than 11.1 mmol/L,and significantly higher than that in the blank group(P<0.05).After 6 weeks of experimental period,the model group was still larger than 11.1 mmol/L,significantly higher than the blank group,the difference was statistically significant(P<0.05),indicating that the difference was statistically significant(P<0.05).The model is successful and stable.After third weeks of intervention,he difference of FBG between the Dioscorea porridge group and the model group was statistically significant(P<0.05).After 4,5 and 6 weeks,the difference of FBG between the Dioscorea porridge group and the model group was statistically significant(P<0.05).2.Comparative analysis of Bifidobacterium in intestinal tract of rats after intervention.Compared with the blank group,the expression of Bifidobacterium in the cecal contents of the model group was significantly decreased,the difference was statistically significant(P<0.05).After the intervention of Dioscorea porridge for 6 weeks,the expression of Bifidobacterium in the content of cecum in the rat congee group increased significantly compared with the model group,and the difference was statistically significant(P<0.05).3.Comparative analysis of intestinal metabolites SCAFs in each group after intervention.Compared with the blank group,the contents of acetic acid,propionic acid and but yric acid in the colon contents of the model group were significantly lower than those in the blank group,and the difference was statistically significant(P<0.05).After the intervention of the Dioscorea porridge for 6 weeks,the content of acetic acid,propionic acid and butyric acid in the colonic contents of the rat congee group increased significantly compared with the model group,and the difference was statistically significant(P<0.05).However,compared with the metformin group,the effect of metformin on acetic acid and butyric acid in the colon content of rats was better than that of the Dioscorea congee group,the difference was statistically significant(P<0.05).4.Comparative analysis of intestinal metabolites LPS in each group after intervention.Compared with the blank group,the plasma LPS level in the model group increased significantly,and the difference was statistically significant(P<0.05).After 6 weeks of intervention,the plasma LPS level in the dioscorea porridge group was significantly 1ower than that in the model group,and the difference was statistically significant(P<0.05).Metformin group had no significant difference compared with the model group,the difference was not statistically significant(P>0.05).Conclusion1.Dioscorea porridge has the effect of adjusting the total bacterial content of Bif idobacterium in T2DM rats and increasing the SCAFs(acetic acid,propionic acid and butyric acid)in the colon of T2DM rats.2.Dioscorea porridge can reduce the level of LPS and FBG in the serum of T2 DM rats.
