| Objective:In this project,the fingerprint of Aralia elata leaves was constructed.L-ooking for the relationship between the“chemical fingerprint spectrum"and the anti-tumor“effect”,the fingerprint data and anti-tumor spectru-m effect relationship of the anti-tumor effective parts of Aralia elata 1eaves is obtained.It provides a reference standard for quality control a nd efficacy evaluation of anti-tumor drugs from Aralia elata species,which can not only clearly define the anti-tumor efficacy component group of Aralia elata leaves,but also lay a foundation for further study on the synergistic combination of its anti-tumor effective components.Methods:1.Establishment and Analysis of Fingerprints of Aralia elata leaves High-performance liquid chromatography(HPLC)coupled with an evapor ative light scattering detector(ELSD)was used to analyze chemical fingerprints of 15 batches of Aralia elata leaves in different regions and a-t different harvesting times;this was promulgated by the National Phar-macopoeia Commission.The“Chinese Medicine Chromatographic Finger-print Similarity Evaluation System Software 2012A Edition”established a fingerprint and evaluated the similarity,using SPSS 20.0 data softwa-re for clustering statistical analysis.2.Anti-tumor activity test and analysisUsing MTT assay,human lung adenocarcinoma cells(A549),human cervical cancer cells(HeLa),human colon cancer cells(HT-29),and human hepatocellular carcinoma cells(SMMC-7221)were used as drug effica-cy models.15 batches of Aralia elata leaves extracts were tested for a-ntitumor activity,using the SPSS 20.0 data software to analyze the obt-ained tumor half-time inhibition rate to obtain IC50 data.3.Anti-tumor spectrum-effect relationship establishmentExcel data software was used to combine the quantitative peak area ofthe fingerprint spectrum with the IC50 of the anti-tumor activity ind ex t-hrough a mathematical model and a gray correlation degree method.Th-e spectrum-effect relationship analysis was performed to construct t he anti-tumor "chemistry spectrum”of Aralia elata leaves-"Pharmaceut ica-1 efficacy" relationship.Results:1.Through the analysis of 15 batches of Aralia elata leaves,the HPLC-ELSD fingerprints of Aralia elata leaves were established.28 fingerprints common peaks were established and the specific components represe-nted by 8 peaks were identified.The similarity between most samples is above 0.9,and the similarity between individual samples is low.2.According to the results of the inhibition rate and IC50 value of 15 total saponins from 4 batches of herbs in this experiment,the anti-tumo-r efficacy of Aralia elata leaves in different areas and at different har-vest periods was somewhat different.3.The mathematical-statistical analysis of the spectral-effect relationshi p-p results showed that there are 12 species of each characteristic comp-onent and the correlation degree of the anti-A549 cell effect contribute-on of 0.8 or more;there are 13 substances that have a correlation deg-ree of 0.8 or more between the 28 characteristic components and the c-ontribution of anti-HeLa cells,and 11 characteristics of the correlation between 28 characteristic components and the contribution of anti-HT-29 cells with a degree of contribution of 0.8 or more;28 substances hav-e a correlation of 0.8 or more with the contribution of anti-SMMC-7221 cells,11 species.Conclusions:1.The HPLC-ELSD fingerprint of total saponins of Aralia elata leaves had 28 common peaks.The relative stability of the saponin components in the leaves of Aralia elata can be reflected,and the method for esta-blishing fingerprints is reliable.The constructed fingerprint map can be used for the quality control of Aralia elata leaves medicinal materials.At the same time,it indicates that the Aralia elata leaves harvested in July,August and September in the northeast of China conform to this standard.2.The anti-tumor activity of Aralia elata leaves from different habitats was different.3.The relationship between HPLC fingerprints and anti-tumor spectrum effect of total saponins of Aralia elata leaves reflects the intrinsic qual-ity of Aralia elata leaves.In the 28 common peaks of the fingerprint,each related peak component has different contribution to different tumo-r cells,and the related peak components show a certain synergistic or antagonistic relationship.The reflected efficacy is the result of the com-bined effects of the relevant components. |
