Effect Of QingHuaYuDu Decoction On Serum Inflammatory Factors And The Expression Of TLR4 Protein In Alcoholic Liver Fibrosis Rats

Objective To study the effect of Qinghuayudu Decoction on the expression of serum inflammatory factors and the expression of TLR4 protein in Alcoholic Liver Fibrosis Rats,to explore the mechanism of Qinghuayudu Decoction in threating Alcoholic Hepatic Fibrosis for providing the theoretical basis of its clinical application.Methods(1)55 SD rats,male,weighing 180g-200g,were randomly divided into blank control group(normal),model control group and Qinghuayudu Decoction group(treatment group),40 animals were raised to establish an animal model of alcoholic liver fibrosis.(2)After the modeling,the treatment group was given the drug(eliminating blood stasis and poison concentration for each kg containing crude drug 1.917g)to high,medium and low group with dose of 200,100,50mg/(kg·D)weight gavage intervention.(3)After the end of the experiment,the ELISA method was used to observe the expression of IL-1β,IL-6,TNF-α.The immunofluorescence and Western-blot method were used to observe the expression of TLR4 in each group.Results(1)The expression of IL-1β,IL-6 and TNF-α,model group was increased than the blank group,and there was a significant difference(P<0.01);Qinghuayudu Decoction groups compared with the model group decreased in high dose group significantly decreased the expression of IL-1β,low dose group can significantly reduce the toxicity and eliminating blood stasis and IL-6 the expression of high dose group and middle dose group can significantly reduce the expression of TNF-α,there were significant differences((P<0.01).(2)The expression of TLR4 protein in the model group was significantly higher than that in the blank group,and there was a significant difference(P<0.01).Qinghuayudu Decoction in high dose group compared with model group decreased,there was a significant difference(P<0.01);And when increase of the concentration of Qinghuayudu Decoction,the decreased expression of TLR4 protein in a concentration dependent manner.Conclusion(1)QingHuaYuDu Decoction can not only decrease the expression of IL-1β、IL-6and TNF-α but also decrease the expression of TLR4 protein.It may be the mechanism for the treatment of alcoholic liver fibrosis by QingHuaYuDu Decoction.
