| Objective:To investigate the effects of the signal transducer and activator of transcription(STAT1)on the interferon-inducible protein NEDD8 ultimate buster 1(NUB1L)expression affecting tumor proliferation ability in hepatocellular carcinoma(HCC).Methods:1.Detect the protein and m RNA expression level of STAT1 in hepatocellular cancer cell HCCLM3,SMMC-7721,Huh7 and normal liver cell line HL7702.2.The plasmid aimed at STAT1 was packed into HCCLM3 and SMMC-7721 cells,and detect the expression of STAT1 protein and m RNA by western blot and q RT-PCR.3.The proliferation capacity of HCCLM3 and SMMC7721 were measured by colony formation assay and Edu assay after regulating STAT1.4.Construct the stably transfected liver cancer cell lines,then detect the proliferation capacity after regulating NUB1 L expression in the stably transfected HCC cells.Results:1.Compared with the normal hepatocellular cells,STAT1 m RNA and protein levels in HCC cells show a relatively low expression state.2.We found NUB1 L is negatively associated with STAT1 expression level.3.STAT1 could negatively regulate the proliferation of HCC cells.4.STAT1 regulated HCC cells proliferation abilities by regulating NUB1 L expression.Conclusion:In hepatocellular carcinoma,STAT1 could regulate HCC cells proliferation by suppressing NUB1 L expression. |
