| Orthogonal design was utilized to optimize the preparing conditions of ethanol extracts of leaves of Rehmannia glutinosa Libosch.The optimum extraction process was to extract three times using tenfold 50%ethanol and 1.0 h per time.The active carbon was utilized to purify the ethanol extract from leaves of Rehmannia glutinosa Libosch.The purification conditions were obtained as follow:active carbon(46 times of ethanol extract)was added to the water solution of ethanol extract(10mg/ml),and placed 12h,then subjected to chromatography column,and eluted with 5BV water and 3BV 30%ethanol successively.The catalpol content of ethanol extract was raised from 6.19%to 15.30%after purification.The macroporous absorption resin was utilized to purify the ethanol extract from leaves of Rehmannia glutinosa Libosch further.The purification conditions were obtained as follow:the water solution of ethanol extract(catalpol concentration:0.5mg/ml)was subjected to H103 macroporous absorption resin at the velocity of 1BV/h,and eluted with 5BV water and 4BV 10%ethanol successively at the velocity of 3BV/h.The catalpol content of ethanol extract was raised from 15.30%to 46.60%after purification.Two acute renal failure mouse models were established,which were induced with aristolochic acid A and glycerol respectively.The anti-acute renal failure effects of ethanol extract from leaves of Rehmannia glutinosa Libosch were studied on the mouse models.The results indicated that the ethanol extract had the effects of anti-acute renal failure mouse models,and catalpol was an active ingredient,and there were other ingredients in ethanol extract from leaves of Rehmannia glutinosa Libosch,which could increase the effects of anti-acute renal failure mouse models in addition to catalpol. |
