Objective: To detect the expression of Borealin protein in colorectal cancer(CRC)and its relationship with other CPC proteins in chromosomal passenger complex(CPC),to explore the effect of down-regulation of Borealin on proliferation,invasion and migration of colorectal cancer.Methods:Immunohistochemistry was used to detect the expression of Borealin,Survivin,Aurora-B and Ki-67 in colorectal cancer tissues.Normal tissues were used as controls to statistically analyze the relationship between the expression of these proteins and the clinicopathological features of colorectal cancer.Highly expressed colorectal cancer cell lines were down-regulated by si RNA to observe the expression of Survivin and Aurora-B in colorectal cancer cells;Western blot,cell proliferation assay,cell scratch assay,immunofluorescence and Transwell assay were used to detect the effects of Borealin/Dasra B down-regulation on the proliferation,invasion and migration of colorectal cancer cells and the expression of related proteins,and further elucidate the role of Borealin/Dasra B in the development of colorectal cancer.Results: The positive expression rate of Borealin in colorectal cancer tissues was significantly higher than that in normal colorectal mucosa tissues(P <0.05),and the expression of Borealin in colorectal cancer was correlated with lymph node metastasis,tumor differentiation degree and depth of invasion(P <0.05).The expression of Borealin was closely related to the expression of Survivin,Aurora-B and the cell proliferation activity marker Ki-67(P <0.01).The LOVO cells with high expression of Borealin were screened by si RNA-borealin in five colorectal cancer cells.The expression of Survivin and Aurora-B was down-regulated after down-regulating the expression of Borealin,which further explained the expression of Borealin.Survivin and Aurora-B were positively correlated.4.Morphological changes after cell transfection,large nuclear multinucleated cells increased,and apoptotic cells increased.The scratch test showed that the healing rate of the tumor cell transfection group was significantly lower than that of the control group;Transwell experiment showed that the number of cells migrated or invaded into the small outdoor group was significantly less than that of the control group.Conclusions: 1.Borealin is highly expressed in colorectal cancer tissues together with Survivin and Aurora-B,and synergistically promotes the development of tumors,and the expression level is positively correlated with the degree of malignancy and invasiveness of tumors.2.Down-regulation of Borealin expression in colorectal cancer cells can inhibit the proliferation,migration and invasion of tumor cells,indicating that it has an important relationship with the occurrence and development of colorectal cancer. |
