Objective: To discuss the influence of salvianolic acid B(SAB)on the proliferation and apoptosis of Human nasal polyps fibroblasts(NPFs).Methods: The NPFs were maintained with SAB at different concentrations.The optimum drug concentration and conditions of salvianolic acid B are determined by CCK-8 assay.The NPFs were divided into four groups: control group which the cells were only cultured in DMEM;the TGF-β1 group which the cells were cultured in 5ng·/ml TGF-β1;the SAB group which the cells were cultured in SAB at the optimum concentration by CCK-8 assay;the TGF-β1+SAB treatment group which the cells were cultured in 5 ng·/ml TGF-β1 and SAB at the optimum concentration by CCK-8assay.The NPFs proliferation of four groups was analyzed by MTT assay.The apoptosis rate of the four groups detected by Annexin V-FITC/PI double staining.The activities of Caspase-3 were determined by the Caspase-3 Kit.Apoptosis-related proteins Bcl-2 and Bax were detected by western blotting.Results: The CCK-8 assay showed that SAB could inhibite the proliferation of NPFs in a dose-dependent pattern.The optimum drug concentration and conditions of salvianolic acid B at 10 μmol/l and the inhibition rate of 72 h was lower than 50% as the best condition.MTT assay showed that SAB could inhibit the proliferation of NPFs stimulated by TGF-β1 in addition.Annexin V-FITC/PI double staining revealed that SAB could induce apoptosis of NPFs,and could inhibit the anti-apoptosis of NPFs stimulated by TGF-β1 in addition.The activities of Caspase-3 were increased by dealing with SAB.Western Blot assessment showed that SAB decreased the Bcl-2protein expression,increased the expression of the Bax protein and improved the ratio of Bax/Bcl-2.Conclusions: Salvianolic acid B has effects on inhibiting proliferation and inducing apoptosis of NPFs.Its mechanism may be associated with the downregulation of Bcl-2 protein,the up regulation of the expression of Bax protein,and increasing the Bax/Bcl-2 ratio and the activity of Caspase-3.
