| ObjectiveChronic periodontitis(CP)is one of the most common oral diseases which causes alveolar bone absorption and tooth loss in adults.In this study,we aimed to remedy chronic periodontitis with Plumbagin(PL),one widely-investigated pharmacologic drug derived from traditional Chinese medical herb.MethodsThe anti-inflammation property of PL is evaluated in TNF-α-induced human periodontal ligament stem cells(PDLSCs)and ligated and Porphyromonas gingivalis(Pg)stimulated Sprague-Dawley rats(SD rats).Proinflammatory cytokines TNF-α,IL-1β and IL-6 were evaluated for analysis of periodontitis pathogenesis.ResultsResults indicated that PL inhibits CP progression by down regulating the expressions of TNF-α,IL-1β and IL-6 in both in vivo and vitro studies(p<0.01).Phosphorylation of the mitogen-activated protein kinase(MAPK),nuclear factor-κB(NF-κB)and JAK/STAT signaling pathways was suppressed after treatment of PL in PDLSCs in western blot(p<0.001).Chemotaxis migration of macrophages was restrained due to reduction of CCL-2 and CCL-5(p<0.01).In the animal experiment,deceleration in bone destruction was observed through Alveolar Bone Loss(ABL)and H&E staining results(ABL: p<0.01).ConclusionWith the given data,conclusion could be made that PL suppressed CP by down regulating the expressions of TNF-α,IL-1β and IL-6,and that the progress was inhibited through MAPK,NF-κB and JAK/STAT signaling pathways in vitro studies,thus indicating its anti-inflammation property against CP. |
