| ObjectiveTo observe the effect of luteolin(Lut)on the growth,apoptosis and autophagy of human tongue squamous cell carcinoma(Tca8113)cells in vitro.MethodsThe effect of Lut on the growth activity of Tca8113 cells in vitro was observed by MTT and plate cloning,according to the experimental results,the suitable concentration and time were selected for subsequent experiments.The apoptosis of Tca8113 cells under Lut was observed by Heochst33342 staining and flow cytometry.Western Blot test further mignonette element of Tca8113 cells c-caspase3 protein,B lymphocyte tumor-2(Bcl-2),the Bcl-2 X protein(Bax)and related Beclin1 protein,LC3Ⅱprotein expression.ResultsThe results of MTT colorimetric method showed that with the increase of Lut concentration,the inhibition rate of cell growth gradually increased,and the IC50 at 24 h was 47μmol/L(P<0.01).The experimental results of plate cloning formation showed that: compared with the control group,the rate of cell cloning formation in the experimental group was significantly slow(P<0.01).At the endof the experiment,the number of clones formed in the experimental group was small(P<0.01),indicating that luteolin could inhibit the growth activity of Tca8113 cells.The results of Heochst33342 showed that the cell apoptosis increased gradually with the prolongation of drug action time(P<0.01).The experimental results of flow cytometry showed that the apoptosis number and apoptosis rate were higher than that of the blank control group after 12 h and24h treatment with luteolin(P<0.01).Western Blot analysis showed that with the increase of drug processing time,the Bcl-2 protein levels were reduced(P<0.05),but increased Bax protein levels,c-caspase3 protein levels(P<0.05),and associated with autophagy Beclin1,LC3Ⅱ expression level also increased(P<0.05).Conclusion1.Luteolin can inhibit the growth activity and induce apoptosis of tongue cancer Tca8113 cells in vitro,which may be related to the activation of bcl-2/Bax/c-caspase-3 pathway.2.Mignonette induced autophagy Tca8113 cells. |
