| Natural products have always been an important source for the discovery of active drugs and lead compounds of active drugs.In modern drug discovery,most synthetic drugs are drugs developed using active natural products as the lead compound.The research of natural products in the discovery of new drugs It occupies an important position in bio-sources,especially microbial-derived natural products,and occupies an important position in the discovery of drugs and lead drugs.These microbial-derived natural products have novel structures and have various good biological activities.In this paper,two fungi from different sources were selected,one from the mangrove plant Kaffonia root sea mud fungus Acremonium citrinum.The laboratory number is MMF4,and the other is the Antrodia cinnamomea symbiotic bacterium Scytalidium cuboideum.The laboratory number is AT6.Strains of fungi were used to study the activity of secondary metabolites.This article uses a variety of separation and purification methods to separate the secondary metabolites of the strain,such as ODS reversed-phase low-pressure chromatography,LH-20 gel column chromatography,HPLC high-performance liquid chromatography,normal phase silica gel column chromatography and other methods.A series of secondary metabolites were isolated from the rice fermentation product of strain Acremonium citrinum.MMF4.At the same time,the structure of these compounds was determined by using spectroscopy techniques such as nuclear magnetic resonance spectroscopy and mass spectrometry.two alkaloid compounds,two diterpene compounds,one cyclopentenol compound,one pyrone compound,two other compounds,of which compounds 2*,3*and 5*are New compounds.A series of compounds were isolated from the fermentation product of the Scytalidium cuboideum.AT6 plate PDA medium.At the same time,the structure of these compounds was determined using spectral techniques,such as nuclear magnetic resonance spectroscopy and mass spectrometry.These compounds include three polyketones Compounds,four diphenyl ether compounds,one alkaloid compound,two other compounds,of which compounds 17*and 20*are new compounds.Using the zebrafish diabetes model to conduct an activity study on the secondary metabolites isolated from strain MMF4,it was found that compounds 4 and 8 can increase the total number of islet β cells at 10 μmol/L,because islet β cells can produce insulin to lower blood sugar Therefore,compound 4 and compound 8 have the potential to lower blood sugar.At the same time,the MTT method was used to detect the cytotoxic effect of the isolated compound on two tumor cell lines.The experimental results found that compound 5*had an effect on BV2 at 20 μmol/L.The cells have moderate cytotoxicity.At 40 μmol/L,they have moderate cytotoxicity on Hela,MCF7,HepG2 and H460 cells.The Juekat-2D10 cell model was used to study the activity of the compound isolated from strain AT6 to reactivate the latent HIV-1 virus.The experimental results showed that compounds 17*and 21 had a certain reactivation effect on the latent HIV1 virus at 10 μmol/L.At the same time,the isolated compounds were tested for MTT cytotoxicity in three tumor cell lines.The experimental results showed that compound 18 and compound 19 had a certain cytotoxic effect on HepG2 and MCF-7 at 20 μmol/L.Compound 21 had obvious cytotoxic effects on MCF-7’and MB231 at 20 μmol/L,and certain cytotoxic effects on HepG2.This study found two compounds with the ability to increase the total number of islet β cells and two compounds with the activity of activating latent HIV-1 virus.A compound with obvious cytotoxic effects on MCF-7 and MB231 was found.The mechanism of action of the compound and its medicinal properties provide a certain material basis. |
