The JAK/STAT signaling pathway is a common pathway for multiple intracellular signaling pathways,such as regulating cell apoptosis,embryonic development,angiogenesis,inflammation,cell proliferation,metastasis,and many other processes.With the development of research,it was found that the JAK/STAT signaling pathway plays a crucial role in the development and progression of many tumors.The JAK/STAT signaling pathway is the key point to regulate cell life activities.Blocking JAK/STAT signaling pathway can inhibit the proliferation of cancer cells and cause apoptosis.OVOL2 protein is a transcription factor with a zinc finger domain and highly conserved in evolution.Recent studies have shown that OVOL2 can inhibit ZEB1 and suppress the occurrence of cancer cell EMT.Thus,it can inhibit the metastatic process of cancer.At the same time,the high expression of OVOL2 protein can reduce the size of mouse orthotopic tumors.This paper revealed the possibility of the interaction between OVOL2 and JAK2/STAT3 signal pathway for the first time.In this paper,OVOL2 overexpresses in breast cancer cell lines.We found that overexpression of OVOL2 significantly inhibited the JAK2/STAT3 signaling pathway and reduced the expression of its downstream genes through real-time quantitative PCR experiments,western blot experiments,fluorescein reporter gene detection and other experiments,.Co-immunoprecipitation experiments further revealed that OVOL2 and STAT3 protein have a binding relationship.At the same time,we monitored the number of cells and detected apoptosis marker proteins.We observed that the high expression of OVOL2 inhibited the proliferation of breast cancer cells and induced apoptosis.When we overexpressed both OVOL2 and STAT3 in breast cancer cells,the degree of apoptosis has decreased.So we believe that OVOL2 can inhibit the JAK2/STAT3 signaling pathway and affect the survival of breast cancer cells.In summary,this article first linked OVOL2 to the JAK2/STAT3 signaling pathway in breast cancer.Meanwhile,we initially revealed the molecular mechanism by which OVOL2 protein inhibits the JAK2/STAT3 signaling pathway and affects cell survival.This finding is of great significance for further exploration of the relationship between OVOL2 and JAK2/STAT3 signaling pathways,as well as the treatment of breast cancer.
