Background and Purpose:Ovarian cancer is the third most common cancer in the female reproductive system worldwide,next to cervical cancer and uterine cancer.With the in-depth study on the pathogenesis and metastasis mechanism of ovarian cancer,the level of diagnosis and treatment of ovarian cancer is gradually increasing.However,due to the deep position of the ovary in the pelvic cavity and the absence of obvious symptoms and signs in the early stage,there is currently no effective way to screen ovarian cancer in the early stage.Most patients with ovarian cancer are already in the advanced stage of the tumor when they seek medical treatment.Moreover,ovarian cancer itself has the characteristics of extensive metastasis in the pelvic cavity and abdominal cavity,which makes the mortality rate of ovarian cancer high and poses a serious threat to the life and health.In recent years,the anti-tumor effect of Astragaloside Ⅳ has become a research hotspot.A large number of literatures reported that Astragaloside Ⅳ can inhibit the proliferation and invasion and migration of different types of tumor cells in vitro.But,there is no research report on whether Astragaloside Ⅳ can inhibit ovarian cancer SKOV3 cells proliferation,migration and invasion.Therefore,the purpose of this experiment is to explore whether Astragaloside Ⅳ can affect ovarian cancer SKOV3 cells proliferation,migration and invasion,and to preliminarily explore its relevant mechanism to provide theoretical and experimental basis for seeking new drugs for the treatment of patients with epithelial ovarian cancer.Methods:Human ovarian cancer SKOV3 cells are treated with different concentrations of Astragaloside Ⅳ(0,0.2,0.4,0.8,1.6,3.2 mg/m L,of which0mg/m L Astragaloside Ⅳ was negative control group).The effects of Astragaloside Ⅳ on the proliferation ability of ovarian cancer SKOV3 cells were investigated by CCK-8 method and plate clone formation experiment.According to CCK-8 results,the concentration of 0.4mg/m L Astragaloside Ⅳ was selected for subsequent tests.The effect of Astragaloside Ⅳ(0.4mg/m L)on the migration and invasion ability of ovarian cancer SKOV3 cells were investigated by Transwell cell migration and invasion experiments.The expression of tumor metastasis-related proteins MMP2 and MMP9 in ovarian cancer SKOV3 cells which treated with 0.4mg/m L Astragaloside Ⅳ for 48 hours was detected by Western Blot,and to preliminarily explore the effect of Astragaloside Ⅳon ovarian cancer SKOV3 cells mechanism.Results: 1 The results of CCK-8 test and plate clone formation test showed that:1.1 Astragaloside Ⅳ can inhibit the proliferation of ovarian cancer SKOV3 cells,with concentration and time dependence.1.2 With different concentrations of Astragaloside Ⅳ treatment of SKOV3 cells,the clone formation rate decreased with the increase of drug concentration,and the clone formation volume decreased with the increase of drug concentration.2 Transwell Cell Migration and invasion experiments showed that :SKOV3 cells treated with 0.4 mg/m L Astragaloside Ⅳ for 24 hours had significantly fewer cells passing through the cells in the migration and invasion experiments than the control group(Astragaloside Ⅳ concentration is 0mg/m L)3 Western Blot showed that:The expression of tumor metastasis-related proteins MMP2 and MMP9 in SKOV3 cells decreased after treated with0.4mg/m L Astragaloside Ⅳ for 48 hours.Conclusion: 1 Astragaloside Ⅳ can inhibit the proliferation of ovarian cancer SKOV3 cells in a concentration and time dependent.2 Astragaloside Ⅳ can inhibit ovarian cancer SKOV3 cells migration and invasion,and its mechanism of action may be related to its down-regulation of the expression of tumor metastasis-related proteins MMP2 and MMP9 in ovarian cancer SKOV3 cells.The above research results preliminarily discussed Astragaloside Ⅳ against ovarian cancer SKOV3 cells,providing theoretical and experimental basis for finding new drugs for the treatment of patients with epithelial ovarian cancer.
