| Objective: To contrast and detect the expression of Carcinoembryonic antigen(CEA)in primary lesions, lymph node metastases, liver metastases and ovarian metastases of colorectal cancer, and compare the expression difference of CEA in primary lesions, liver metastases, lymph node metastases and ovarian metastases of colorectal cancer and analysis the potential significance. Methods: From January 2014 to December 2019 45 patients with advanced colorectal cancer treated in the Cancer Medical Hospital of Guangxi Medical University were retrospectively selected, about 40 primary lesions were collected(including 24 samples with liver metastases,16 samples with ovarian metastases),about 54 paired specimen of metastases(including 26 lymph node metastases,15 liver metastases, and 13 ovarian metastases)from those 45 were collected.35 samples with both primary lesions and different metastases site, about 13 samples including between primary lesions and two metastatic were received at the same time(including 9 samples primary lesions with lymph node metastases and liver metastases,4 samples have matched primary cancer with lymph node metastases and ovarian metastases)in 35 samples, about 22 samples including between primary lesions and one metastases were received at the same time(including 11 samples primary lesions with lymph node metastases,5 samples primary lesions with liver metastases, and 6 samples primary lesions with ovarian metastases)in 35 samples.Primary lesions, lymph node metastases, liver metastases and ovarian metastases of colorectal cancer of CEA expression was detected by immunohistochemistry.Based on staining depth score in cancer tissue + staining range score,0 to 1 points was Negative "-",2~3 was weak positive "+",4~5 was positive "++",6 was strong positive "+++".Among them,(-)and(+)was divided into negative,(++)and(+++)was divided into positive,(CEA positive cases /(CEA negative cases + positive cases))× 100% = positive rate. The data obtained in this experimental study was analyzed using SPSS 25.0 software.Rank data was tested using Wilcoxon rank sum test,P<0.05 was considered statistically significant.Results:1.The positive rate of CEA in 40 primary lesions of colorectal cancer was97.5%(39/40),the positive rate of CEA in 26 lymph node metastases was84.6%(22/26),the positive rate of CEA in 15 liver metastases was 93.3%(14/15),the positive rate of CEA in 13 ovarian metastases was 92.3%(12/13).There was no statistically significant difference in the expression of CEA between primary lesions and lymph node metastases(P>0.05).It was no statistically significant difference in the expression of CEA between primary lesions and liver metastases(P>0.05).There was no statistically significant difference in the expression of CEA between primary lesions and ovarian metastases(P>0.05).2.In the 24 matched lymph node metastases and primary lesions, the positive rate of CEA in primary lesions was 100.0%(24/24),and the positive rate of CEA in lymph node metastases was 83.3%(20/24),There was no statistically significant difference in the expression of CEA between paired primary lesions and lymph node metastases(P>0.05).3.In the 14 matched primary lesions and liver metastases, the positive rate of CEA in primary lesions was 92.9%(13/14),the positive rate of CEA in liver metastases was 92.9%(13/14).There was no significant difference in the expression of CEA between paired primary lesions and liver metastases(P>0.05).4.In 10 matched primary lesions and ovarian metastases, the positive rate of CEA in primary lesions was 100.0%(10/10),and the positive rate of CEA in ovarian metastases was 100.0%(10/10).There was no significant difference in the expression of CEA between paired primary lesions and ovarian metastases(P>0.05).5.The positive rate of CEA in primary lesions of colorectal cancer was97.5%(39/40),the positive rate of CEA in primary lesions of colorectal cancer with liver metastases was 95.8%(23/24),the positive rate of CEA in primary lesions of colorectal cancer with ovarian metastases was 100.0%(16/16).There was no statistically significant difference in the expression of CEA between the primary lesions of colorectal cancer with liver metastasis and the primary lesions of all colorectal cancer(P>0.05).There was no significant difference in the expression of CEA between primary lesions of colorectal cancer with ovarian metastasis and primary lesions of all colorectal cancer(P>0.05).There was no significant difference in the expression of CEA between the primary lesions of colorectal cancer with ovarian metastasis and the primary lesions of colorectal cancer with liver metastases(P>0.05).Conclusion: The results of this study show preliminary that CEA is highly expressed in primary lesions, lymph node metastases, liver metastases and ovarian metastases of colorectal cancer.CEA may not be suitable as an indicator for the detection of cell heterogeneity in different parts of colorectal cancer. |
