Objective:To study the oxidative stress ability and mechanism of photoaging keratinocytes model enhanced by the water extract of Acronychia peduncμlata,which provided theoretical basis for developing Zhenzhen perfume to improve skin photoaging.Methods:1.HaCaT cells were treated with different concentrations of the water extract of Acronychia peduncμlata.The proliferation rate of HaCaT cells was determined by cck-8 method,screened out the effective and safe concentrations for HaCaT cells.2.Irradiate the cells with UVB rays of 30,60 and 90m J/cm~2,and screened out the semi-lethal radiation dose.3.The cell model of photoaging HaCaT was divided into groups.The blankgroup received no intervention of UVB radiation and the water extract of Acronychia peduncμlata;The model group received no intervention of UVB radiation but with the water extract of Acronychia peduncμlata;The experimental group received intervention of UVB radiation and tthe water extract of Acronychia peduncμlata.4.Using prophase the water extract of Acronychia peduncμlata’s safe concentration interference light aging HaCaT cell model,detection before and after the intervention HaCaT cells reactive oxygen species(ROS)and malondialdehyde(MDA)content,superoxide disproportionation enzyme(SOD),catalase(CAT),glutathione peroxidase(GPx)and antioxidant enzyme activity and total antioxidant capacity(T-AOC)level changes.Results:1.The cck-8 test resμlts showed that:compared with the blank group,4mg/L-64mg/L concentration of the water extract of Acronychia peduncμlata coμld significantly inhibit the proliferation of HaCaT cells,and the proliferation rate was significantly reduced(P<0.05).The concentration of0.5mg/L-2mg/L coμld promote the proliferation of HaCaT cells,and the proliferation rate was significantly increased(P<0.05).The concentration of0.25mg/L had no significant effect on cell proliferation,and the difference was not statistically significant(P>0.05).Therefore,0.5mg/L-2mg/L was selected as the safe and effective concentration in this experiment.2.When the radiation dose reached 60mj/cm~2,the relative activity of the cells was 53%,reaching the semi-lethal state.60mj/cm2 was used as the radiation dose for the follow-up experiment.3.Compared with the blank group,the proliferation rate of cells in the model group and the experimental group were significantly reduced(P<0.05).But the proliferation rate of cells in the experimental group was significantly higher than the model group(P<0.05).4.Compared with the blank group,ROS and MDA contents in cells of model group were significantly increased(P<0.05),SOD,CAT,GPx activity and T-AOC level were significantly reduced(P<0.05);After intervention with a concentration of 2mg/L reductive perfume extract,MDA content in cells of experimental group was lower than that in blank group(P<0.05),SOD activity was not significantly different from that of the blank group(P>0.05);After intervention with other concentrations(0.5mg/L,1mg/L)of healthy fragrance extracts,the ROS and MDA contents in cells of the experimental group were all higher than those in the blank group(P<0.05),SOD,CAT,GPx activity and T-AOC levels were all lower than the blank group(P<0.05).However,compared with the model group,ROS and MDA contents in of the experimental group were significantly reduced(P<0.05),SOD,CAT,GPx activity and T-AOC level were significantly increased(P<0.05).Conclusion:A certain concentration(0.5-2mg/L)of healthy fragrance extract can improve the defense ability of photoaged keratinocytes against UVB radiation.By removing ROS in photoaged HaCaT cells,reducing MDA content,and increasing the activity of SOD,CAT,GPx and other antioxidant enzymes and t-AOC level in photoaged HaCaT cells,the antioxidant stress ability of HaCaT cells can be improved.It may be one of the important mechanisms by which the extract of Healthy fragrance promotes the anti-light damage of HaCaT cells.
