| Objective: Downregulating the expression of Livin through shRNA to explore the sensitivity of Cal 27 to 5-fluorouracil(5-FU),thus to provide some help to solve the chemotherapy resistance of oral cancer.Method: The designed shRNA sequence targeting Livin were constructed to lentivirus and infected Cal 27 to get the stable Livin-silencing cell strain.After treated with 20μM,40μM,60μM 5-fluorouracil to 24 h,the cell apoptosis and cell viability were measured by flow cytometry and MTT respectively.Results: The lentivirus of designed shRNA targeting Livin were successfully constructed.After silencing Livin,the sensitivity and cell apoptosis of Cal 27 to 5-FU were elevated dramatically,and the cell viability decreased significantly(P<0.05).Higher expression of pro-Caspase 3 was observed in the Livin-silencing group.Conclusion: The expression level of Livin was downregulated in Cal 27 after transfected with shRNA,which increased the sensitivity of Cal 27 to 5-FU.The underlying mechanism is related to the block of activation of Caspase 3. |
