OBJECTIVES To investigate the underlying mechanism of HP1α regulating SUZ12 in the developement of cholangiocarcinoma.METHOD TCGA datasets were used to detect the expressions of HP1α and SUZ12 in cholangiocarcinoma samples.Westernblot and RT-PCR were used to detect the expression of HP1α and SUZ12 in clinical biospecimen and cells.Transfection with lentivirus were used to construct stable regulation of HP1α and SUZ12 in cholangiocarcinoma cell line Hucct1.Transwell assay,CCK8 assay and Edu assay were used to detect the migration,invasion and proliferation of cholangiocarcinoma cells.Westernblot and Co-immunoprecipitation were used to detect the regulation of HP1α to SUZ12 and the expression of HP1α/SUV39H1/DNMT3A/H3K9me3 complex and CDKN2 A.RESULTS HP1α and SUZ12 were upregulated in cholangiocarcinoma.Downregulation of HP1 and SUZ12 repressed the cell proliferation but not the migration and invasion.Downregulation of HP1α decreased SUZ12 but not EZH2 and EED.HP1α regulated the expression of CDKN2A/INK4 A by direct interaction with SUZ12 but not the HP1α/SUV39H1/DNMT3A/H3K9me3 complex.CONCLUSION HP1α regulates CDKN2A/INK4 A expression by interaction with SUZ12 in cholangiocarcinoma proliferation.
