Objective: Osteosarcoma is the most common primary solid malignant tumor of bone,and infestation is the main biological characteristics.Hypoxia-inducible factor(HIF)under hypoxic microcirculation is closely related to the infestation of osteosarcoma.This article aims to explore the role of HIF-3α in the infestation of osteosarcoma,and to provide a new treatment strategy for the treatment of osteosarcoma.Methods:(1)Immunohistochemical staining was used to detect the expression of HIF-3α in osteosarcoma tissue.(2)Using semi-quantitative RT-PCR and Western blot molecular biology methods to detect the effect of hypoxia on the expression of HIF-3α in MG-63 cells.(3)Cell adhesion experiment,cell invasion experiment and cell invasion experiment were used to detect the effect of hypoxia on cell adhesion,migration and invasion ability of MG-63 cells.(4)After knocking down HIF-3α in MG-63 cells by transfecting HIF-3α si RNA,the effects of normoxic and hypoxic conditions on cell adhesion,migration and invasion ability were detected.(5)After transfection of HIF-3αplasmid to overexpress HIF-3α in MG-63 cells,the effects of normoxic and hypoxic conditions on cell adhesion,migration and invasion ability were detected.Results:(1)Immunohistochemical results showed the high expression of HIF-3α in osteosarcoma tissue.(2)Hypoxia promoted the expression of HIF-3α m RNA and HIF-3αprotein in MG-63 cells.(3)Hypoxia enhanced the adhesion,migration and invasion ability of MG-63 cells.(4)The knockdown of HIF-3α in MG-63 cells reduced the ability of adhesion,migration and invasion.(5)The overexpression of HIF-3α in MG-63 cells enhanced the ability of adhesion,migration and invasion.Conclusion: Hypoxia promotes the expression of HIF-3α in osteosarcoma MG-63 cells,which in turn enhances the invasion and metastatic ability of the cells,indicating that HIF-3α plays an important role in the invasion and metastasis of tumor cells caused by hypoxia.
