BACKGROUND:Circular RNA(circRNA)is a new class of endogenous non-coding RNA that plays a crucial role in various types of tumors.However,information on the involvement of circRNA in the treatment of gastric cancer(GC)remains limited.Our previous study showed that circ-EIF4G3 is obviously up-regulated in GC tissues compared with adjacent tissues,and can promote the proliferation,invasion and migration of GC cells.In this study,the specific mechanism of circ-EIF4G3 promoting gastric cancer proliferation,invasion and migration will be further explored.METHODS:Microarray microarray was used to detect BGC-823 cells in the si-circ-EIF4G3 and si-NC groups,and a group of significantly differentially expressed miRNA(miRNA)were found.Using circinteractome bioinformatics software-assisted analysis,it was found that circ-EIF4G3 may interact with miR-335.The binding sites of circ-EIF4G3 and miR-335 were confirmed by luciferase reporter gene and fluorescence in situ hybridization(FISH).The interaction between circ-EIF4G3 and miR-335 was studied by transfection of miR-335 mimics.Results:1.Nuclear separation and FISH results show that circ-EIF4G3 is mainly located in the cytoplasm;2.Using microarray chip technology to detect BGC-823 cells in si-circ-EIF4G3 and si-NC groups,found a group of significantly differentially expressed miRNA,including miR-335;3.Luciferase reporter gene experiments showed that circ-EIF4G3 has a binding site with miR-335;4.circ-EIF4G3 promotes proliferation,invasion and migration of GC cells by sponge adsorption with miR-335.The results of CCK8,Transwell and Scratch experiments showed that the cell proliferation,invasion and migration ability of the si-circ-EIF4G3+miR-335 mimic treatment was significantly inhibited compared to the GC cells treated with si-circ-EIF4G3.Conclusion:Our study demonstrates that circ-EIF4G3 promotes proliferation,invasion and migration of GC cells in the cytoplasm by adsorption of miR-335.
