Objective: The purpose of this study was to study the expression level of lysyl oxidase-like proteins-1 in central nervous system gliomas and its correlation with clinical parameters and to explore the possible mechanism of promoting tumor progression.Method:In this study,RNA sequencing of 749 glioma patients with complete clinical and survival data were downloaded from the CGGA and included in the study.Another RNA sequencing expression data with clinical and survival data were downloaded from the TCGA as the validation cohort.According to the median value of LOXL1 expression level,it was divided into high expression group and low expression group,clinical correlation analysis,prognosis analysis and related pathway analysis of LOXL1 in different types of glioma were analyzed.Forty-three patients with intramedullary glioma admitted to the Department of Neurosurgery of the Union Hospital of Fujian Medical University from February 2012 to February 2017.Immunohistochemical method was used to detect the expression level of LOXL1 in spinal cord intramedullary gliomas,and the survival prognosis was analyzed based on the survival data.Through the bioinformatics analysis of GO,GSEA,KEGG,etc.,the biological processes related to the LOXL1 gene are explored to understand the possible mechanism of promoting tumor progression.Result:1.In the CGGA and TCGA databases,the expression of LOXL1 was positively correlated with the WHO grade and IDH mutation of glioma(p <0.05).At the same time,the overall survival rate of patients with high expression of LOXL1 was significantly lower than that of patients with low expression of LOXL1(p <0.05).Univariate and multivariate COX analysis shows that LOXL1 can be clinically used as a predictor of independent poor prognosis of glioma.2.The expression of LOXL1 was positively correlated with the WHO grade of glioma in clinical samples of related intramedullary gliomas in our hospital(p <0.05),and the overall survival rate of patients with high expression of LOXL1 was significantly lower than that of patients with low expression of LOXL1 Patients(p<0.05).3.Bioinformatic analysis suggests that LOXL1 may promote the progression of glioma by affecting the biological functions of extracellular matrix remodeling,angiogenesis,intercellular adhesion and negative regulation of T cells.Conclusion:The expression level of LOXL1 is positively correlated with the WHO classification of gliomas.It shows a higher expression in gliomas with a higher degree of malignancy,which is a more reliable predictor of malignant gliomas.Patients with high expression of LOXL1 have a poor clinical prognosis,which is an independent prognostic factor for glioma.In addition,LOXL1 may promote the progression of glioma by affecting biological behaviors such as extracellular matrix remodeling,angiogenesis,intercellular adhesion,and negative regulation of T cells.In summary,we can conclude that LOXL1 can be used as an indicator of the survival prognosis of central nervous system gliomas,and it may also become a new therapeutic target.
