| Objective: To observe the effect of Pectic Polysaccharides(PP),extracted from Rauwolfia verticillata(Lour.)Baill.var.hainanensis Tsiang,in Dextran sulphate sodium(DSS)induced ulcerative colitis in mice.The observed contents included:general condition,DAI score,local histopathology of colon,expression of TNF-α and IL-6 in local tissue,and expression of MAPKs in local tissue.According to the results,explore the effect and mechanism of this method on UC.Methods: A total of 80 BA-8/C female mice,aged 6-8 weeks and weighing17-22 g,were randomly divided into(1)normal control group,(2)DSS group,(3)DSS + SASP treatment group,(4)DSS + PP treatment group.After the experiment,the mice in each group were observed daily mental state(whether excited or fatigued),good,Appetite(whether good,better than normal,poor),hair(gloss,thickness,shedding,vertical hair),nutritional status(obesity,normal,weight loss,dyscrasia),daily weighing and record,the diarrhea and bloody stools of the mice were observed,fecal traits(normal,semi-soft,and soft)were recorded,and fecal occult blood was measured using the benzidine method,and the disease activity index score(DAI)of the mice was evaluated.On the 8th day,modelsmaking is finished,the normal control group and the DSS model group were intragastrically administered with 0.2 m L of0.9% saline once a day,two of drug intervention group receive gavage with PP(100mg/kg)and SASP(100 mg/kg)respectively.And continued to observe and score.Each group of mice was sacrificed on day 15.Colonic mucosal tissue(inflammation)was taken for HE staining.The location and expression of ERK,p38 MAPK and JNK protein in colonic mucosa of each group were detected by immunohistochemistry and Western blotting.The expression of TNF-αand IL-6 in colon tissue of each group was detected by ELISA.Results:1.General conditions in mice: The gastrointestinal symptoms,fecal traits,alertness,hair,appetite status,and activity of the SASP-treated and PP-treated mice were significantly better than those in the DSS model group.2.DAI scores: At the end of the experiment,there was a significant difference in DAI between the SASP treatment group and the PP treatment group compared with the model group(P<0.05).3.Histopathology: SASP and PP treatment group mice with colonic mucosal injury and submucosal edema and inflammation were significantly reduced,histological changes were significantly improved.4.The expression of MAPKs in colonic mucosa of each group was detected by immunohistochemistry: Compared with DSS model group,the expressions of ERK,p38 MAPK and JNK protein in SPSA and PP treatment group decreased significantly(P<0.05).The results showed that SASP and PP can inhibit the expression of ERK,p38 MAPK and JNK protein in mice UC inflammatory cells.5.The expression of p-ERK,p-p38 MAPK and p-JNK protein in colon tissue of each group was detected by Western blot: Compared with the DSS model group,the expression of SPSA and PP treatment group protein was decreased(P<0.05),and the difference was statistically significant.It is suggested that the SPSA and PP treatment groups have anti-inflammatory effects.6.Effects on the expression levels of UC inflammatory factors TNF-α and IL-6:Compared with the DSS model group,the levels of TNF-α and IL-6 in the colon tissue of the SPSA-treated group and the PP-treated group were significantly lower,and the PP-treated group had Statistical significance(P <0.05).7.The levels of TNF-α and IL-6 in colon tissue were determined by ELISA:Compared with the DSS model group,the levels of TNF-α and IL-6 in the colon tissue of SPSA treatment group and PP treatment group were significantly lower than those in the PP treatment group.(P<0.05).Conclusion:1.Pectic Polysaccharides(PP),extracted from Rauwolfia verticillata(Lour.)Baill.var.hainanensis Tsiang,can improve the symptoms of bloody stool and diarrhea in UC mice,relieve its pathological damage,and promote the healing of colonic mucosal injury.2.Pectic Polysaccharides(PP),extracted from Rauwolfia verticillata(Lour.)Baill.var.hainanensis Tsiang,achieves treatment of colitis in DSS mice by intervention of MAPK Signaling Pathway,which down-regulates the expression of ERK,p38 MAPK and JNK. |
