| Objective: To investigate the significance of activating transcription factor-2(ATF-2)expression in diffuse large B-cell lymphoma,not otherwise specified(DLBCL,NOS),and to analyze the relationship between the expression of ATF-2 and clinicopathological features,as well as prognosis of DLBCL,NOS.Methods: Collectting 60 cases of DLBCL,NOS and 40 cases of lymph node RH.Detectting the expression of ATF-2,Cyclin D1,BCL-2,CD10,BCL-6,MUM1 protein in DLBCL,NOS cases and the expression of ATF-2 protein in lymph node RH cases using immunohistochemistry staining.The expression of the above protein in DLBCL,NOS and the expression of ATF-2 protein in lymph node RH were observed by light microscopy.To group DLBCL,NOS cases by the Hans classification principle(GCB group and non-GCB group).Comparing the expression of ATF-2 protein in DLBCL,NOS and lymph node RH by statistical methods.Analyzing the relationship between the expression of ATF-2 protein and the prognostic grouping,the expression of BCL-2、Cyclin D1 protein,clinicopathological features in DLBCL,NOS.Results:(1)Immunohistochemical staining showed that ATF-2 protein expressed in nucleus both of DLBCL,NOS and lymph node RH cases,which showed different immunostaning intensity.45 cases showed high expression of ATF-2 protein and 15 cases showed low expression of ATF-2 protein in all 60 cases of DLBCL,NOS;16cases showed high expression of ATF-2 protein and 24 cases showed low expression of ATF-2 protein in all 40 cases of lymph node RH.The expression of ATF-2 protein was significantly difference between the DLBCL,NOS cases and lymph node RH cases(P = 0.000).(2)Immunohistochemical staining showed that there was no significantly difference of expression of ATF-2 protein in different age,gender,Hans classification of DLBCL,NOS cases(P>0.05).(3)Immunohistochemical staining showed that the expression of ATF-2 protein has no relationship with the expression of Cyclin D1 and BCL-2 protein in DLBCL,NOS(P >0.05).(4)In the 60 cases of DLBCL,NOS,45 cases of DLBCL,NOS with high ATF-2 protein expression has 8cases died of disease which has an average survival time of 5.75±3.88 months;15cases with low ATF-2 protein expression has 6 cases died of disease which has an average survival time of 16.67±17.08 months.Kaplan-Meier survival analysis showed there was no significantly difference of overall survival between the high ATF-2 protein expression and the low ATF-2 protein expression in DLBCL,NOS cases(P=0.194).Conclusion:(1)ATF-2 protein expressed in nucleus both in DLBCL,NOS cases and lymph node RH cases.(2)The expression of ATF-2 protein in DLBCL,NOS was significantly higher than that in lymph node RH.Speculating the high expression of ATF-2 protein is related to the occurrence and development of DLBCL,NOS.(3)The high expression of ATF-2 in the nucleus of DLBCL(NOS)can provide a new idea for studying its pathogenesis and clinical treatment. |
