With the recent environmental deterioration(such as the increase in air haze,exhaust and dust mites from automobiles),the incidence of asthma is also increasing.Therefore,it is of great significance that we further explore the pathogenesis of asthma and discover the drugs which can treat asthma.Diacerein,Diacetyl dihydroanthracenecarboxylic acid,is an important inhibitor of osteoarthritis,clinically has anti-inflammatory and analgesic pharmacological effects.However,whether it is useful in treating inflammatory diseases remains unknown.The purpose of this study is to explore the specific mechanism of Diacerein in relaxing airway smooth muscle(ASM)in mice.This study could provide some ideas for finding medicines of asthma.Firstly,at the organizational level,force measuring system was used to explore the effect of drugs on contraction of isolated tracheal rings.We found that Diacerein can relax high-K~+-or ACh-induced contraction of tracheal ring in a dose-dependent manner.These contractions can be blocked by Nifedipine,Pyr3 and Gadolinium.Therefore,we wondered whether Diacerein can inhibit the currents of channels such as VDCC and NSCC?Secondly,in order to further explore its mechanism of action,patch-clamp systems were performed to record relevant ion channel currents at the cell level.Our data show that Diacerein blocks the currents in the VDCC,NSCC plasma channels.Finally,on the living body level:(1)Pulmonary function meter was used to detect respiratory resistance(Rrs)caused with ACh,we found that Diacerein can obviously reduce this resistance.(2)Asthma model was constructed,we found that asthma symptoms were improved well in mice treated with medication by HE、PAS and RT-PCR analysis.Take together,these data suggest that Diacerein has great potential for treating asthma.
