| Callicarpa nudiflora is a plant belonging to the genus Callicarpa of the verbena family.Its dry aerial parts are used as medicinal herbs and it has the effects of hemostatic and analgesic,antibacterial and anti-inflammatory,loosening blood stasis and swelling,expelling wind and dampness,and detoxifying.The prescription or proprietary Chinese medicine made from it is used to treat various types of bleeding disorders such as acute infectious hepatitis,gynecological inflammation,burns,scalds,bleeding from the digestive tract,and bleeding from trauma.It is a native medicine from Hainan.In this paper,the chemical components of the dichloromethane extraction site of Callicarpa nudiflora ethanol extract were systematically separated,and the anti-cervicitis activity of Callicarpa nudiflora ethanol extract was preliminary studied.In this project,the dichloromethane fraction of Callicarpa nudiflora ethanol extract was studied.The ethanol-heated reflux extraction method was used to extract the fat-soluble components of Callicarpa nudiflora to obtain the alcohol extract of Callicarpa nudiflora,and then extracted with petroleum ether,dichloromethane,and n-butanol in order to obtain each extraction site.Finally,the dichloromethane portion of the Callicarpa nudiflora alcohol extract was separated by silica gel column chromatography,Sephadex LH-20 gel column chromatography,and semi-preparative high-performance liquid phase.As a result,six lipophilic components were obtained,two of which were triterpenoids,two were flavonoids,one was sesquiterpene lactone compounds,and one was an unsaturated fatty acid compound.According to the analysis of physical and chemical constants,chemical methods and spectral data,the structures of the compounds were identified,and the six compounds were identified as: α-amyrin(1),5,4′-dihydroxy-3,7,3′-trimethoxyflavone(2),5-hydroxy-3,7,3′,4′-tetramethoxyflavone(3),Oleic acid(4),oleanolic acid(5),loliolide(6).Among them,compounds 4 and 6 were isolated from this plant for the first time.This project conducted a preliminary study on the anti-cervicitis activity of Callicarpa nudiflora.A cervicitis model in rats was replicated by intravaginal administration of phenol glue,The blank group,model group,Callicarpa nudiflora alcohol extract high-dose group,Callicarpa nudiflora alcohol extract low-dose group,and positive drug group were given different types and doses of therapeutic drugs.During the subsequent treatment,observe and record the relevant data every day,take the rat cervical tissue for observation,prepare sections,and analyze by HE staining.The results showed that the method of replicating rat cervicitis model with phenol glue was simple,high success rate and good reproducibility.Callicarpa nudiflora alcohol extract has a significant therapeutic effect on cervicitis in rats,and has no significant effect on the weight and organ index of rats during the treatment process.The cervical tissue of the rats was dissected and observed.Compared with the model group,the rats in the high-dose group,the low-dose group,and the positive control group had significantly reduced vaginal redness and swelling,and the vaginal mouth secretions significantly reduced or disappeared.The vaginal and cervical tissue edema was significantly improved;Histopathological examination revealed that the glandular glands in the cervical tissue of rats in the low-dose group and high-dose group of Callicarpa nudiflora extract basically returned to normal,inflammatory cell infiltration was significantly suppressed,no epithelial cell degeneration and necrosis,and tissue hyperemia was significantly relieved Vascular congestion has also improved significantly. |
