Objective: To elucidate the pathogenesis of PCOS phlegm-dampness syndrome from the NF-κB inflammation pathway,and to explore Qigong Pill’s effectors related to PCOS phlegm-dampness syndrome-mediated NF-κ B inflammation pathway——P-IκBα,IKKβ,lipopolysaccharide(LPS)and inflammation The influence of factors TNF-α,IL-6,IL-1β is expected to provide a new target for the treatment of PCOS phlegm and dampness syndrome by traditional Chinese medicine,and provide strong theoretical and clinical support for this field.Method:t is planned to select PCOS patients and normal endocrine patients(control group)who were treated in the Reproductive and Genetic Center of Shandong Provincial Hospital of Traditional Chinese Medicine from January2019 to January 2020 as the research object,including 20 patients with PCOS phlegm and dampness syndrome,and PCOS There were 20 patients with phlegm and dampness syndrome and 20 patients in the control group.The height and weight of patients in each group were measured,and the body mass index(BMI)was calculated.The chemiluminescence method was used to detect the sex hormone index,fasting blood glucose(FPG),and serum fasting insulin(FINS)in each group.The insulin resistance index(HOMA-IR)was evaluated by a steady state model);Detection of TNF-α by chemiluminescence immunoassay;Detection of serum levels of IL-6,IL-1 β,LPS,IKK β,P-I κ B α in each group by enzyme-linked immunoassay(ELISA);and analysis of IKKβ and P-IκBα,Correlation of TNF-α,IL-6,IL-1β,LPS and FPG,FINS,HOMA-IR.At the same time,patients with PCOS phlegm-dampness syndrome were treated with Qigong Pill Fang,and the improvement of TCM syndromes and BMI,FPG,FINS,HOMA-IR,TNF-α,IL-6,IL-1β,LPS,IKKβ,and P were observed before and after treatment.-IκBα changes.Results:1.General situation and comparison of hormone levels: serum LH,T,LH / FSH values in PCOS group were significantly higher than those in control group(P <0.01);PCOS phlegm and dampness group had high BMI,FPG,FINS,HOMA-IR values In the non-phlegm-dampness group and the control group,the difference was statistically significant(P <0.01).2.Comparison of inflammatory factors TNF-α,IL-6,IL-1β and LPS,IKKβ,P-IκBα levels:PCOS two groups of TNF-α,IL-6,IL-1β and LPS,IKKβ,P-IκBα levels They were significantly higher than the control group(P <0.01),and the levels of TNF-α,IL-6,IL-1β,LPS,IKKβ,and P-IκBα in the PCOS phlegm and dampness group were higher than those in the non-phlegm and dampness group.P <0.05 or P <0.01).3.Correlation analysis: The overall serum IKKβ level of the study subjects was positively correlated with P-IκBα,TNF-α,IL-6,IL-1β,LPS,FINS,HOMA-IR(P <0.05).4.The clinical efficacy of Qigong Pills in treating PCOS phlegm and dampness syndrome: After treatment,the BMI,FPG,FINS,HOMA-IR level and TCM syndrome scores of patients in PCOS phlegm and dampness group were all improved.Academic significance(P <0.01 or P <0.05).5.The effect of Qigong Pills on the levels of TNF-α,IL-6,IL-1β and LPS,IKKβ,P-IκBα: after treatment,the level of each factor was lower than before,the difference was statistically significant(P <0.01).Conclusion:1.PCOS patients have chronic inflammation,and the performance of PCOS phlegm dampness group is more obvious;2.PCOS Phlegm-Dampness Syndrome patients have obvious characteristics of insulin resistance;3.Dry-dampness-phlegm prescription Qigong Pill may reduce serum IKKβ,P-IκBα,LPS,TNF-α,IL-6,IL-1β levels,reduce i NFlammation,thereby improving insulin resistance in patients with PCOS and phlegm-dampness syndrome to achieve therapeutic purposes.In summary,it is speculated that TNF-α,IL-6,IL-1β,and LPS may mediate the process of insulin resistance by up-regulating the expression of IKKβ and P-IκBα related molecules in the i NFlammatory signaling pathway,and participate in the development of PCOS phlegm dampness syndrome.
