Prunella vulgaris L.is a very common Chinese herbal medicine in clinical treatment of traditional Chinese medicine.It has the the clinical effects of clearing liver fire,clearing eyes,dissipating tubercles and dispersing knots.Granula Prunellae and Electuary Prunellae have similar functions with L.in curing diseases mentioned above.They are often used as an alternative of Prunella vulgaris L.,because they are easy to carry and to be dosed.The quality standards of Prunella vulgaris L.and its single prescription preparations are difficult to evaluate their quality comprehensively due to the unitarization of quality indicators and the loss of determination specificity.Today,fingerprint technique,as a comprehensive and quantifiable detection method,has now been widely used and accepted in the quality control of TCM.Using the chromatographic fingerprint to assess the batch-to-batch consistency of chemical composition and the determination of those bioactive compounds is widely applied both in China and the world.Therefore,the content of 3 major phenolic acids including salviaflaside(the characteristic principle),rosmarinic acid,caffeic acid was simutiniously determined,and then the HPLC fingerprint was investigated using the same determination condition.This study provided a comprehensive analysis method for interior quality control of Prunella vulgaris and its single prescription preparations.Experiments as follows were accomplished:(1)High performance Liquid chromatography was employed to determine three principle active constituents simultaneously in Prunella vulgaris L.and its single prescription preparations.Baseline separation was performed well on the phenomenex Luna C18 Column with Gradient Eution of acetonitrile and 0.5%(V//V)glacial acetic acid as mobile phase.The results indicated that three principle components(salviaflaside,rosmarinic acid,caffeic acid)were separated well under the analytical condition.Good linear correlation,precision,stability,reproducibility and recovery were presented.(2)High performance liquid chromatography with diode array detector was employed for fingerprint analysis of Prunella vulgaris L.and its single prescription preparations on the phenomenex Luna C18 Column with Gradient Eution of acetonitrile and 0.5%(V/V glacial acetic acid as mobile phase.The results indicated that Prunella vulgaris L.and its granule have more common peaks and they can obtain a good similarity and quality consistency in batch-to-batch.On the contrary,some batches of Decoction Prunella are diferent from the standard spectrum.Principle component anlysis(PCA)helped us to find some samples from the group,and the peaks with high contribution to the PCA were revealed.Through this study,we can evaluate furtherly the quality of Prunella vulgaris L.and its single prescription preparations in a reasonable and effective way.It can help us to solve the defect that the quality indicator of current quality standards is monobasic,and to find blind spots and pain points of drug supervision.The quality indicators and test methods established in this study can help us to find the matters such as Inferior materials being sold as superior materials,substitution materials being used in single prescription preparations of Prunella vulgaris L.,and can help us to break the hidden rules of the industry.Focusing on the prominent problems and potential risks,we will improve the quality of medicines constantly,and ensure the safety and effectiveness of medicines for the public.
